ObjectiveIn the mechanism of the balloon and nylon loop-induced arterial injury and hypertrophy of inner membrane,the injury of the smooth muscle cells in the medial membrane is the key.Impaired smooth muscle cells release basic FGF which roles in the other vascular smooth muscle cells in the way of Paracrine and Induces the emergence of a variety of growth factor,resulting in the proliferation of vascular smooth muscle cells.This study was to use balloon and nylon loop injurying rat carotid artery model to track arterial wall changes with time after mechanical damage and explore the importance of membrane damage in the formation of hypertrophy of inner membrane.MethodsWistar rats(male,12 weeks,220 g)are choosed,24 in balloon group,24 in nylon loop group,8 in control group.2Fr balloon and 4-0 nylon loop injury the rats's carotid artery,rats were killed in batches on 6h,12h,24h,2d,3d,5d,7d,14d after injury,Dyeing organizations and Immunohistochemistry were preformed on the injury artery,Observation was gived to the damage arterial wall,detect BrdU-positive cells appear rate,the emergence of PCNA-positive cells and the rate of migration of cells in the inner membrane and medial membrane.ResultsThere were almost no BrdU-positive cells in the control group,there were some same points between balloon group of nylon loop,that is,the medial smooth muscle cells significantly increased,inner membrane thickening,BrdU-positive cells and PCNA-positive cells index of injuried artery manifests a biphasic increase with 2 peaks. There were PCNA-positive migrational cells after injury.Different points,in the various indicators of Observation and determination,Nylon loop was significantly weaker than balloon group.Conclusion2Fr balloon and 4-0 nylon loop were inserted in the carotid artery,Stripping the endothelial,at the same time resulting in medial membrane injury,BrdU-positive cells index of injuried artery manifests a biphasic increase with 2 peaks in the second day and the fifth day.There was positive correlation between the medial membrane damage degree and inner membrane hypertrophy.so in the forming process of inner membrane hypertrophy,the medial membrane injury plays an important role.
