Background:The development of cell immunobiology and molecular immunobiology promotes immunological application, which is one of the fundamental methods in tumor treatment.Currently adoptive cellular immunotherapy(ACI) is popular . Cytokine-induced killer cell is a new type of immunological killer cell, which is one of the hottest in ACI research.At present research about carcinoma -related immunological cell and clinical data indicate that cytokine-induced killer cell is an efficient and new MHC-unlimited immunological cell with tumor-killing ability of wide spectra.,which has high speed of hyperplasia ,high killing ability , little adverse clinical effects,and so on.CIK cell is one of the most efficient immunological cell in tumor killing.Section I The proliferation profile in vitro and anti-tumor activity of CIK cells from healthy persons and patient with primary liver cancerObjective To Observe the differences of CIK(cytokine induced killer)cells from from healthy persons and patient with primary liver cancer , in their morphology, phenotype, proliferation activity and cytotoxic activity.Methods CIK cells were generated by IFN-γ,CD3McAb,IL-2,IL-1 induction of culturing peripheral blood mononuclear cells (PBMC) of both 6 healthy blood donors and 6 liver-cancer patients. These effective cells were identified at different intervals by flow cytometry analysis. The proliferation activity of these effective cells were determined. MTT assays were used to measure the cytotoxic activity on SMMC-7721 cells in vitro.Results 1.There was no significant difference in proliferation performance of CIK cells from between healthy persons and liver-cancer patients. (p<0.05) CDC cells have the abilities of easy culture and quick proliferation. 2. There was no significant difference in killing activities on SMMC-7721 of CIK cells from between healthy persons and liver-cancer patients. (p<0.05) But the killing function on SMMC-7721 are remarkable. 3. There was no significant difference in DC-specific costimulatory molecules of CIK cells from between healthy persons and liver-cancer patients. (p<0.05) But following the culturing-time goes on ,both caused an increase in corresponding cell surface markers such as CD3+CD56+,CD8+ and an decrease of CD45RA+ on CIK cells.Conclusion CIK cells from two sources of healthy persons and liver-cancer patients both have the ability of easy culture ,quick proliferation and high quality of killing rate of liver-cancer cells. There was no significant difference.Section II The anti-tumor effects of CIK cells combined with chemotherapyObjective To determine the changes of CIK cells in their phenotype, proliferation activity, cytotoxic activity in vitro and then further investigate the assist therapeutical effect of CIK cells.Methods CIK cells were generated by culturing PBMC of healthy blood donor. Then the changes in the proliferation activity and phenotype of the cells were determined. MTT assays were used to determine the cytotoxic activity in vitro. The anti-tumor activity of CIK cells were evaluated after chemotherapy in Balb/c nude mice bearing SMMC-7721 liver cancer.Results CIK has the assistant treatment function to the chemotherapy, after treats 25 day, CIK cell to tumor inhibitory action obvious strong in independent chemotherapy group.Conclusion CIK cells could enhance the anti-tumor effect of chemotherapy remarkbly.
