Objective To investigate the expression of hypoxia-inducible factor-1α(HIF-1α) and E-cadherin and Snail proteins in rectal carcinoma and their significance.Methods: We retrospective analysis the 101 rectal cancer patients'clinicopathologic data from Jan 2001 to Dec 2002 of our hospital who had undergone radical surgery, and detect the expression of HIF-1αand Snail and E-cadherin proteins by using immunohistochemical SABC method in these rectal carcinoma tissues.Results: The positive rate of HIF-1αin rectal cancer was 72.3%and the Snail is 78.2%, and the E-cadherin was 37.6%. The expression of HIF-1αand E-cadherin were significantly related with the tumor differentiation and lymph node metastasis and Dukes'stage of rectal cancer (P<0.05); The expression of Snail was significantly related with the lymph node metastasis and Dukes'stage of rectal cancer (P<0.05).The expression of Snail is significantly negative correlation with that of E-cadherin(r=-0.332,P<0.01); the expression of HIF-1αis significantly negative correlation with that of E-cadherin(r=-0.432,P<0.01); the expression of HIF-1αis significantly positive correlation with that of Snail(r=0.612,P<0.05).The expression of HIF-1αand Snail and E-cadherin were correlation with the rectal cancer patients'survival rate after accepted the radical surgery(P<0.05);each of the Dukes stage and lymphonodus-metastasis and the expression of HIF-1αand E-cadherin and snail is an independent prognostic predictor in patients.Conclusions The over-expression of HIF-1αand Snail, and the decreased expression of E-cadherin might be important biological marker for malignant transformation and invasion and metastasis of rectal carcinoma.The expression of HIF-1α, E-cadherin and Snail has positive effects on predicting the prognosis in rectal cancer.
