The anti-tumor potential of components of Chinese herbal medicines has attracted widely interests. Alisol B acetate isolated from Alismatis rhizome has the bioactivity of inducing apoptosis of several cancer cells and reversing the sensitivity of multi-drug resistant cancer cells to anti-tumor drugs. Besides of Alisol B acetate, Alismatis rhizome consists of a great deal of triterpenes with the similar molecular configuration which may have the same bioactivity. Thus the extraction and separation of Alismatis rhizome and the bioactivity of these constituents were studied in this paper.Three extraction and separation methods were taken in this study including traditional way of diacolation - organic solvent extraction - column chromatography, preparative thin layer chromatography, and supercritical CO2 extraction. Series of mixtures were obtained through these methods and then analyzed by thin layer chromatography or HPLC. Advantages and disadvantages of these methods were discussed in this article.In order to determine the inhibitory effects of constituents from Alismatis rhizome on SP2/0 cancer cells, O.D. value was detected at the wavelength of 490nm with MTT colorimetry. The results indicated that the IC50 values of 3 ingredients were below 100μg/mL, and the lowest IC50 value was 74.60μg/mL, which was still higher than the standard IC50 value for anti-tumor mixture of herbal medicines.( IC50<40μg/mL for mixture). The inhibitory effect of E-2 ingredient, which had been proved to be the most effective inhibitor of SP2/0 cells in this study, was further detected through morphological methods. In conclusion, some constituents of Alismatis rhizome have inhibitory effects on SP2/0 cancer cells depending on their concentration and time, but still can not be used as anti-tumor drug.
