Objective: To explore the neuroprotective mechanisms of r-HuEPO on rats after traumatic brain injury.Methods: 60 healthy adult male Sprague-Dawley rats were randomly divided into 5 groups: 1000U/kg r-HuEPO-treated, 3000U/kg r-HuEPO-treated, 5000U/kg r-HuEPO-treated, nicholin and normal saline (control group) groups. The model was made by improved Feeney's traumatic brain injury model. Serum samples were collected at 6h, 24h, 3d, 5d, 7d after trauma. The serum S100B protein and IL-6 content of each group after treatment was detected with double antibody Sandwich ELISA.Result: (1)Changes of the serum S100B protein content: compared with the saline control group, the level of S100B protein in 1000U/kg r-HuEPO-treated group was not significant difference (P>0.05), but it was significant lower in 3000U/kg r-HuEPO-treated and 5000U/kg r-HuEPO-treated groups (P<0.001). Compared with the nicolin control group, the serum S100B protein content increased significantly in 1000U/kg r-HuEPO-treated group(P<0.001), but it in 3000U/kg r-HuEPO-treated and 5000U/kg r-HuEPO-treated groups were not significant difference(P>0.05). Compared with the 1000U/kg r-HuEPO-treated group, the serum S100B protein content was significant lower in 3000U/kg r-HuEPO-treated and 5000U/kg r-HuEPO-treated groups(P<0.001), and it was more lower in 5000U/kg r-HuEPO-treated group than it in 3000U/kg r-HuEPO-treated group(P<0.05); (2) Changes of the serum IL-6 content: compared with the saline control group, the serum IL-6 content was significant lower in each group(P<0.001). Compared with the nicolin group, it was not significant difference between 1000U/kg r-HuEPO-treated and 3000U/kg r-HuEPO-treated groups(P>0.05), but it was higher in 5000U/kg r-HuEPO-treated group(P<0.05). Compared with the 1000U/kg r-HuEPO-treated group, the serum IL-6 content was lower in 3000U/kg r-HuEPO-treated group (P<0.01), and it was not significant difference in 5000U/kg r-HuEPO-treated group (P>0.05). And the serum IL-6 content was significant lower in 3000U/kg r-HuEPO-treated group than it in 5000U/kg r-HuEPO-treated group (P<0.01).Conclusion: (1) r-HuEPO can significantly cut the serum S100B protein content down in rats after traumatic brain injury, and the decrease degree of it was positive correlated with the dosage of r-HuEPO. (2) r-HuEPO can significantly cut the serum IL-6 content down in rats after traumatic brain injury, and the medium dosage r-HuEPO decreased it more lower than others. (3) Inhibition of the composition and secretion of S100B protein and IL-6 by EPO might be one of the mechanisms in decreasing inflammatory reaction in system and partis. And based on it, EPO may educe its neuroprotective effect after traumatic brain injury.
