| Objective:Investigating the effect of toxin factors on human germ cells by human sperm survival assay to test the cultures material used in IVF laboratory.Methods:Using two types of culture wells to test the effect on sperm of ovum aspiration needle,embryo transfer catheter,surgical gloves and 1ml injection needle,and to compare with the survival rate of sperm through washing and non-washing of ovum aspiration needle, embryo transfer catheter and surgical gloves.Results:The survival rate of sperm of Nunclon's culture dish is 0.63,higher than Falcon's culture dish 0.50(x~2=6.8762 P=0.0087 P<0.01);No difference was found between standard and ovum aspiration needle,embryo transfer catheter,BD injection needle and Jie-rui injection needle (x~2=1.4493 x~2=0.1642 x~2=0 x~2=0.0102,P>0.05);Compare the survival rate of sperm of washing and non- washing of ovum aspiration needle,embryo transfer catheter and surgical gloves,the outcome is F=2.329,F=0.029,F=0.655 respectively(P>0.05);No different was found between the survival rate of sperm of BD injection needle and of Jie-rui injection needle(F=0.655,P>0.05).Comparison the survival rate of sperm among ovum aspiration needle group and embryo transfer catheter group and injection needle group,there were no significant differences.Comparison the survival rate of sperm of ovum aspiration needle group and embryo transfer catheter group and injection needle group with surgical gloves group respectively,the differences were statistically significant.Conclusion:Bioassay toxin of Nunclon's culture dish is lower than Falcon's culture dish;We cannot wash the ovum aspiration needle,embryo transfer catheter and surgical gloves before using them;surgical gloves has significant bioassay toxin, washing cannot decrease the bioassay toxin;The bioassay toxin of Jie-rui injection needle made in China is similar with BD injection needle made in U.S.A. |
