Objective: To improve the preparing method of extra-thins lice of human preimplantation embryos.Methods: The three pronuclear (3PN) human preimplantation embryos were fixed at 2-8 cell stage. We used Agar to pre-embed and then got the extra-thins lices.Results: Even the ultra-structure of few embryos can be observed with this method.Conclusion: Agar pre-embed method is an efficient method to observe the ultra-structure of few embryos. Objective: To establish the detection method of apoptosis in human preimplantation embryos by TdT-mediated nick end labeling.Methods: The three pronuclear (3PN) human preimplantation embryos were fixed at 2-10 cell stage. TdT-mediated nick end labeling (TUNEL) and laser confocal scanning microscopy were used to detect apoptosis signs in embryos.Results: No positive TUNEL signals were found in normal and arrested 3PN embryos. The TUNEL assay was positive in 20% of fragmented embryos. The positive signals were green fluorescence which was observed in cell nuclei.Conclusion: TUNEL is an effective method to detect apoptosis in human preimplantation embryos.
