Part one: The Distribution of Ca2+ in Acupionts And the Effect of Acupunting on the Ph ValueExperimentⅠ:Experimental Study on the distribution of Ca2+ in urinary bladder meridian of goatsObjective: To explore the relation of calcium ion and meridian phenomenon.Methods: To observe the distribution characteristic of Ca2+ real-timely anddynamically,in acupoints of urinary bladder meridian and in no-acupoints of goats with the needle-type calcium sensor.Results: The concentration of calcium ion in acupoints is higher than that in no-acupoints (p<0.01 or p<0.05), which suggest that the enrichment Phenomena of calcium ion exist in meridian.Conclusion: Calcium ion may be the capital agent in acupuncture signal transduction, the determination of concentration of calcium ion could be regarded as the important physical and chemical marker in identifying the acupoints. ExperimentⅡ:Effect of Acupunting on the Ph Value of Acupoints of GoatsObjective:To investigate the relationship between acupuncture and the pH value of acupoints and non-points .Methods :Measuring the pH value of acupoints and nonpoints with the pH sense needles at the same time and separately.Results :1 The pH value descends when acupunturing ,and there are statistical differences between the extent of descending of acupoints and nonpoints ;2 When acupunturing synchronously there is no obvious difference between them .Conclusion :1 Acupunture may increase the metabolism of points more obviously ;2 The local metabolism influences each other when acupunturing synchronously.Part two: Effect of Acupuncture on the VEGF Expression and Regional Blood Blow in the Rats with Cerebral IschemiaObjective: to study the change of regional cerebral blood flow(rcbf) in the rats with cerebral ischemia after acupuncted 3 times,and detect the expression of vegf-mrna ,vegf and cell proliferation.To provide the theoretic guidance for investigating the mechanism of the protection of acupuncting for cerebral ischemina.Methods:1. 36 healthy male SD rats were chosen for experiment ,which divided randomedly for normal ,model and experiment groups. A rat model of acute cerebral ischemia was built by photochemically initiated thrombosis .2. cerebral blood flow measurementchanges in cerebral blood flow(CBF) at the surface of the cortex were recorded using a laser-Dpller flowmetry monitor equipped with a small caliber probe of 0.4mm in diameter.Rats were anesthetized with Chloral Hydrate.Once anesthetized, the rats were placed on stereotactic.A midline incision was made.The scalp and periosteum were reflected.Three points were marked to ensure consistent measurements.①A point,0.5mm lateral to the sagttal suture and 1mm posterior to the coromal suture on the left side;②B point,3mm to the sagttal suture which overlies the ischemic core region;③C point,5mm lateral to the dagittal suture on the left side which overlies the perifocal region. 3 Half of the rats were killed after infracted 72h.The brains of which were removed rapidly and frozen at -70℃for RT-PCR.Another half were fixed with polyformadehyde for immunohis tochemistry.Results:1 In RT-PCR vegf-mrna expressions in acupuncted group are obviously higher than others.2 In immunohistochemistry,the positive granules of vegf and ki67 are more in acupuncted group than model group.3 The rcbf in acupuncted is also increased in C point more than others.The rcbf in A point is not changed in every group,which in B point is decreased in every group.Conclusions:1 Acupuncting can increase the rcbf in perifocal.2 Acupuncture increasing the rcbf in perifocal maybe correlate with the increasing of vegf protein and cell proliferation.
