Influenza virus is a kind of virus coated with the envelope.Influenza virus A often outbreak and cause worldwide pandemic.In history,each influenza pandemic tireaten human health and cause economic losses.The reason is that surface antigen produce variation.So,it is necessary to study the vial exterior fine structure.Now, we investigate influenza virus A(H1N1)by AFM.Partâ… Objective;Through observing the morphology of the prepared influenza virus (H1N1)with transmission electron microscopy(TEM)and atomic force microscopy AFM)to explore the application of AFM on the research of the external character of viruses.Methods;TEM image was obtained by negatively stained influenza virus with 1% Phosphotungstic Acid;AFM image applies the tapping mode to influenza virus without any further treatment in air at room temperature,and the topographic feature parameters,including length,diameter,Ra and Rq are calculated by sectional analysis.Results;The shapes of influenza virus A observed by AFM and TEM have many forms,such as spherical,filamentous and so on.TEM image of influenza virus-A is two-dimensional image,and viral surface has visible spikes,and AFM exhibits the three-dimensional image that can be described with several quantifiable indexes through sectional analysis.AFM phase images show viral surface clearly which is characterized by rugged feature and gear-like protuberance.Partâ…¡;Objective;AFM investigate the modality of influenza virus A treated with detergent.To compare the destructive effect on viral external structure of various detergents and select an effective detergent applied to dissolve viral membrane structure.Methods;AFM observation the morphology of viruses treated with NP-40 and EDTA.Results;Breakages of viral structure are very different when viruses are treat with different concentration NP-40.The higher of the concentration of NP-40, the severer of viral external structure destroyed.But EDTA is less valid than NP-40 and it is not regular. Partâ…¢;Objective;Through investigation the modality that integrated viruses and viruses treated with NP-40 that combine with three mouse sera to discuss the effect of NP-40 that damage the surface structure of the virus and impact the interaction of the virus and mouse sura and to explore atomic force microscope as a tool of the type A influenza virus immune detection method.Methods;TEM and AFM investigate the morphology of integrated virus and the viruses treated with NP-40 and the morphology of viruses combined with three mouse sera,and measure the topographic feature parameters,including length,diameter,Ra and Rq are calculated by sectional analysis.Results;AFM images show the topography of different viruses and the viruses combined with three mouse sura and the topographic feature parameters. AFM images reveal that the sera changeviral size result in the diameter increase. Nevertheless,TEM images shows that the intergrated viruses combined with H1N1 immune mouse sera appear a phenomenon of agglutination.And the viruses combine with H1N1 and FN protein immune mouse sera also show a phenomenon of agglutination.To compare the viruses treated with NP-40 to integrated viruses that combined with three sera,NP-40 damages viral surface structure and affects interaction of antigen and antibody,and the roughness of viral surface increases,but the viral size is not changed.So,the roughness can be a criterion that NP-40 damages viral surface structure and affectsactivity of antigen and antibody.And the viral diameter can be an immunological index of atomic force microscopy.
