Millettia nitida Benth.var. hirsutissima, is a kind of traditional Chinese herbs, is used as medicine with long history, and is a characteristic traditional Chinese herb of Jiangxi. In the Chinese medicine, it has efficiency of blood tonic to invigorate the blood and activating meridian used to treat menoxenia, blood deficiency sallow, numb paralyse, rheumatism numbness and so on. We preliminarily studied enrichment, constituent anlysis and antioxidant activity of flavonoids from Millettia nitida Benth.var. hirsutissima Z.Wei in this thesis. It will offer the theoretical foundation for popularization and clinical practice of Millettia nitida Benth.var. hirsutissima. The thesis divides four chapters to carry on the elaboration, its main contents are as follows:In first chapter, a review was presented, in which introduced plant origin, the progress of chemical constituent of Caulis Spatholobi, clinical application, the methods of extraction and separation and analytical of natural product flavonoids.In the second chapter,the extracts of total flavonoids was determined by UV,and choose the standard sample which contain daidzein, genistein, isoliquritigenin, formononetin and biochanin to analyse the extracts of constituents by HPLC. Compare the methods of extract and sepration which is better.In the third chapter,we choose macroporous adsorption resins to enrich the total flavonoids of Millettia nitida Benth.var. hirsutissima. Through static experiment and dynamic experiment,we can optimized the better process parameters which the macroporous adsorption resins enriched the total flavonoids. The results show that the two resin of AB-8 and H103 had good capability in adsoption capacity and desorption rate,and the total flavonoids was enhanced one time at list after adsorped by macroporous adsorption resins. adsorption contrention,adsorption pH,eluents, adsorption and desorption rate,are the important factors which effected the macroporous adsorption reson adsorped the flavonoids.In the forth chapter, we choose the standard sample BHA and BHT to compare with the extracts of Millettia nitida Benth.var. hirsutissima. in antioxidant research. The model are 1,1-Diphenyl-2-picrylhydrazyl (DPPH) radical scavenging activity, Superoxide scavenging activity, Hydroxyl radical scavenging activity, Test for ferric ion reducing capacity (Fe3+ to Fe2+), Test for ferrous ion chelating activity (binds Fe2+), Protective effect on Fenton reactant-induced DNA sugar damage. The results show that the extracts have different effects on scavenging free radical activities directly or indirectly .The EtoAc extract has the best activity, Chloroform extract take after,petroleum ether extract has the worst activity,and the other extracts activity quite. The extracts activity have closely relationship with total flavonoids, especially the flavone structure and content. EtoAc extract and Chloroform extract can instead the synthesis antioxidant of BHA and BHT,as nature antioxidants.
