To report a new method for the establishing spontaneously metastatic animal model of human esophagus carcinoma and the metastatic-lymph node-deriving subline, meanwhile, to study the characters of subline derived from human esophagus carcinoma cell line ECa-109.Methods: Eca-109, human esophagus carcinoma cell line, was injected into the gastric mucosa layers of SCID mouse with a concentration of 1-2×10~6 in 0.2 ml solution. All mice were sacrifed 3 months later or when the mice were dying. The described mice were investigated pathologically, part of the metastastic lymph node were inoculated subcutaneously for the establishment of metastatic subline. A new subline was established using primary tissue culture and the characters of the subline were determined by MTT assay, werstern blot, Transwell method and wound-healing method.Results: The primary cell was obtained and named NMC109 cells. The proliferation of the NMC109 cells was increased, the migration and invasiveness potential of the cells was enhanced.Conclusion: It suggested that the method could employ to obtain high metastatic potential cells.
