Galic acid(GA), 3-4-5-tri-Ortho-Hydroxybenzoic Acid, is existed in plants, such as grape, tea. it is a kind of multi-phenolic compounds in nature. GA has many biologic activities such as anti-inflammatory, anti-mutation, and anti-oxidation . Objective: To study the protective effect and mechanism of GA on myocardial ischemia-reperfusion injury.Methods:Rats were randomly individed into four groups: control group ,model group ,DS group and GA group .Control and model groups rats were injected saline i.v. by the dose of 2ml/kg ;DS group rats were injected DS injection by the same dose and route;GA group rats were injected GA i.v. by the dose of 10ml/kg .After the left anterior desending coronary artery (LAD) ligated for 40 min, then reperfused for 120 min.The following indexed were evaluated : the change of ST segment(J point) of ECG , the activity of SOD, the expression of Bcl-2, Bax and iNOS detected by immunohistochemistry..Results: Compared with control group rats , the ST segment of ECG was elevated in rats of model ,DS and GA groups ( P<0.01),GA can alleviate the raising of J point ( P<0.05). Compaed with control group rats , the content of MDA was increase obviously in model rats ,and the activity of superoxide dismutase (SOD) was decreased(P<0.01), GA can alleviate the chang of SOD and MDA induced ischemia–reperfusion injury. GA can decrease the experession of iNOS, up-regulate the expression of Bcl-2 and down-regulate Bax , therefore inhibit the apoptosis of myocardium cell , as well as.decreasing the myocardium injury. Conclusion: GA can protect myocardium against ischemia-reperfusion injury with anti- apoptosis mechanism.
