Ovarian cancer is common in female. Becuase of the deep location of ovary in pelvis and no efficient early diagnosis method, 60-70% patients are found to be advanced ovarian cancer patients. The death rate of ovarian cancer is the highest among all kinds of female genital system cancers. Main therapy methods for ovarian cancer are operation, chemotherapy and radiation therapy. Although there have already been a lot of improvement on these therapy methods, the long-time survival of ovarian cancer patients is not prolonged too much. Therefore, it is very important to find better early diagnosis method and new therapy method.Prostatic acid phosphatase (PAP) is synthesised by prostatic epithelial cells, which is mainly distributed in the prostate and prostate cancer. The PAP levels in serum, caused by PAP-secreting tumor cells, are elevated in patients at all stages of prostatic carcinoma, and PAP was suggested to be a more accurate indicator of micrometastatic disease compared with the Gleason score and the prostate-specific antigen (PSA) level. In addition, PAP is a promising target molecule in specific immunotherapy for metastatic androgen-independent prostate cancer. For many years, The tissue distribution of PAP has been believed to be restricted to the prostate and prostate carcinoma. Although there have been some reports showing that PAP is expressed in tissues other than the prostate and prostate cancer, few systematic investigations about the PAP expression in non-prostate tissue or tumor have been carried out. We have also reported that PAP is expressed in colon, breast and gastric adenocarcinoma and that PAP-specific cytotoxic T lymphocytes (CTLs) can show cytotoxicity to these tumor cells. In this study, This experiment examines the expression level of PAP in the ovary cancer cell and the tumor , so as to provide experimental basis of ovarian cancer's shift and recrudescence.1. The expression of PAP mRNA in ovarian carcinoma cell linesThe expression of PAP mRNA in various ovarian carcinoma cell lines was investigated by the RT-PCR method. Total RNA was isolated from 1×10~7 ovarian carcinoma cells, cDNA was synthesized by RT-PCR. PAP cDNA was detected by PCR amplification using a set of oligonucleotide primers specific to PAP. The mRNA expression of PAP was detected in 2 (HO-8910 and ES-2) of 4 ovarian adenocarcinoma cell lines(HO-8910,HO-8910pm,ES-2 and SK-OV-3).2. The expression of PAP protein in ovarian carcinoma cell linesThe expression of the PAP protein in ovarian carcinoma cell lines was examined by Western blot analysis. The cytoplasmic protein of different ovarian carcinoma cell lines were extracted according to the manual. The protein was separated by SDS-PAGE. The proteins in the acrylamide gel were blotted to a Hybond-polyvinylidene difluoride membraneand probed with Anti-PAP mouse monoclonal antibody. The PAP protein was found to be expressed in ES-2 .3. Expression of PAP in ovarian cancer tissues The PAP expression of ovarian adenocarcinoma tissues were examined by immunohistochemical staining. As a result, 3 ovarian cancer tissues out of 6 serosity corpora mammillaria ovarian cancer tissues were weakly positive for PAP.In conclusion, expression of PAP mRNA and PAP protein in ovarian cancer cell lines and tissues were examined. PAP mRNA and PAP protein were revealed to be expressed in ovarian cancer cell lines. PAP protein were weakly positive in in situ ovarian cancer. PAP could be involved in the metastasis and recurrence of ovarian cancer.
