Effect Of Endostatin On The Expression Of Bcl-2,Bax In Nude Mice Implanted With Human Breast Cancer Cell

ObjectiveTo study the effect of endostatin on the expression of Bcl-2,Bax in nude mice implanted with human breast cancer cell.Methods1.Model simulating human breast cancer was established by hypodermic implantation of histologically intact tumor tissue into nude mice.2.Endostatin was administered sc at dose of 20.0mg/kg,10.0mg/kg and 0mg/kg every other day for seven weeks.3.Eight weeks after implantation,Bcl-2,Bax,apoptotic index were evaluated respectively after the mice were sacrificed.4.Using SPSS 12.0 sofeware analyses the data,and gets conclusion.Result1.Compared with the untreated control,growth of the tumor was significantly reduced in size in mice treated with endostatin with an inhibition of rate 0, 58.4%,91.3%at the dosage of 0mg/kg,10.0mg/kg and 20.0mg/kg, respectively.2.The MVD was also decreased significantly in the treated mice(9.6±1.2) versus(4.6±0.9),(1.8±0.4).The growth of breast cancer implanted in nude mice were significantly inhibited(P＜0.05).3.The incidences of liver(lung) was also significantly inhibited in the treated mice at dose of 20mg/kg(P＜0.05).4.The AI of tumor cell was increased significantly,in the treated mice (3.50±0.94)%,(8.29±2.60)%,(17.46±4.50)%,(P＜0.05).5.Expression of Bcl-2 in tumor cell was significantly reduced in mice treated with endostatin with gray scale(0.41±0.12),(0.70±0.21),(0.90±0.10) at the dosage of 20.0mg/kg,10.0mg/kg and 0mg/kg(P＜0.05).6.But the expression of Bax in tumor cell didn't show significantly distinction in three teams with gray scale(0.51±0.10),(0.50±0.10),(0.55±0.09), (P＞0.05).7.The experiment showed Endostation changed proportion of Bcl-2 and Bax in tumor cell and stimulated tumor cell to apoptosis.ConclusionsOne mechanism of Endostation stimulating tumor cell to apoptosis is downregulation expression of the pro-survival protein Bcl-2.Mesh endostatin;nude mice;breast cancer;Bcl-2;Bax...
