Background: Gastric cancer (GC) is the malignant tumor derived from stomach mucosal epithelium , accounting for 9 percent of malignant tumors from stomach, and 95 per- cent of stomach cancers each year in the total number of new cancer cases. Especially, the mortality rate of gastric cancer have been occupying the forefront of the world in China, Japan, Vietnam and other East Asian countries because of the habits of diet, climate and geographical impacts, the incidence rates of gastric cancer and mortality have also raised in recent years. The incidence of gastric cancer in China's high rate of cancer mortality of the first, the average mortality rates as high as 20/100 thousand. Highly metastatic gastric cancer is a result of the main causes of death, the early detection of gastric cancer metastasis-related genes to prevent its occurrence and development, prognosis and treatment plays an important role in gastric cancer at this stage of basic research is a hot one.Objective: Select the metastasis-related genes in gastric cancer with high invasion risk and discuss the probable molecule mechanism of pathway in the process of gastric cancer. The establishment of Reverse Transcription Loop-mediated Isothermal amplification rapid detect high-invasive gastric cancer metastasis-related genes.Methods: Three paires of metastasized to live tissue and carcinoma in situ tissue of stomach, all of which were men and in the same stage of age, were used as experimental group and control group respectively to establish high-flux cDNA expression spectrum chips. First chose the up-regulating or down-regulating genes simultaneously in all the three chips. The software of Pathway v1.0 was applied in the investigation of multi-kinds molecular pathways which may influence the genesis and progression of stomach carcinoma in the already selected genes. RT-PCR was employed to determine the transcription of mRNA to the related genes in several cases of metastasis stomach carcinoma in the primary and metastasis tissues. Gastric cancer samples with liver metastasis were extracted RNA, then amplified the metastasis-associated gene-Sepine1 by Loop-mediated Isothermal Amplification(LAMP). The condition is remained 30min in 63℃and its amplification products were identified by endonuclease.Results: The up-regulating CLDN1 gene and down-regulating UGT1A10 gene selected using the pathway analysis in this study can completely accord with qualitical detection of 6 cases by RT-PCR. RT-LAMP can rapidly amplify the specifical gene.Conclusions: The pathway analysis can find molecule pathways and related-genes on the basis of the primarily selected correlation genes in the chip, providing the evidences and references for the genes study for gastric cancer with metastasis. RT-LAMP can be widely applied to tumor metastasis-associated gene detection, early diagnosis of clinical standards because it is specifical, rapid and efficient advantage.
