Objective:①To contrast the determination of propofol biological samples after pretreated with the methods of protein precipitation,liquid-liquid extraction,solid-phase extraction and detected with HPLC-UV;②To investigate the effection of storage temperature and light condition to propofol biological samples'stability;③To survey the effection of the combination drugs on the determination of propofol.Methods:①To pretreat propofol biological samples with the methods of protein precipitation, liquid-liquid extraction,solid-phase extraction after improvemented and detected with HPLC-UV.To inspect HPLC chromatography conditions.The Symmetry(?) C 18(4.6mm×250mm 5.0μm) column and acetonitrile-waters(70:30)as a mobile phase were used.The detective wavelength was 272 nm.The flow rate was 1.0ml/min.The column temperature was 35℃;②To determinate whether the combination drugs will impact on the content of propofol by the change of the measured concentration and the joined concentration with fluorescence method and HPLC method.Results:①The three kinds of methods show that the linear range were 0.1-10μg·ml-1.The limit of detection was 50ng·ml-1(S/N=3).The method of protein precipitation have a low recovery rate and more impurity extration, steep baseline,bad peak.The method of liquid-liquid extraction is equal with the solid-phase extraction by the recovery rate.But the former has a great interference in extract material background,many impurity peak and the environment pollution is severe; the latter can acquire sincere sample,but high costs;②The content of propofol is on descending trend in varying degree at room temperature,4℃and serial freezing temperature.Natural lighting and Shade condition as same as above;③Diazepam, Ketamine,Atropine,Fentanyl have not UV absorption.The measured concentration of propofol is different and the joined from the numerical point when the concentration series Tramadol was added.Conclusion:①From extraction method itself,HPLC map,the recovery rate and the treatment consideration of whole blood,solid-phase extraction method is better than the former two;②4℃is the feasible preservation temperature to the propofol plasma sample.But the light was little effect on it;③Diazepam,Ketamine, Atropine,Fentanyl have not impact on the determination of propofol.To Comparison the measured concentration and the joined by t-test.Then conclud that the concentration series Tramadol will not influence on the determination of propofol when detected with fluorescence and HPLC.
