Objective:Through observating changes of IL-1β?IL-10 mRNA expression and protein concentration in the myocardial tissue of rats with oleic acid-induced acute respiratory distress syndrome ,and study the effect of IL-1β?IL-10 during myocardic injury. Methods: The ARDS model were established in rats by intravenous oleic acid (OA) via tail vein. Thirty-two Wistar rats were randomly divided into 4 groups: control(C)?OA2h?OA4h?OA6h. Lung wet/dry weight, the pathomorphologic changes of myocardial tissue and serum myocardic enzymes (AST, CK, CK-MB, LDH) were observed in each group. The method of radioimmunoassay (RIA) was adopted to determine the IL-1βand IL-10 concentration in serum and myocardial tissue. The expression level of IL-1β?IL-10 mRNA in myocardial tissue was detected by Semi–quantitative RT-PCR. Results: After oleic acid injection, the lung W/D in OA group were significantly higher than control(P<0.01). Compared with control group, serum AST increased(P<0.01). Serum level of LDH in OA2h, OA4h were significantly increased than control group (P<0.01), while there was no statistical difference between OA2h and OA4h(P>0.05). Serum level of CK and CK-MB in OA2h?OA4h group had increased compared with the control group (P<0.05), and the increase of OA2h was higher than OA4h(P<0.01). The histopathological change of myocardial were observed in OA group:with various degrees, congestion, edema, inflammatory cell infiltration and necrosis. The IL-1βlevel of OA6h in serum and myocardium were obviously higher than control group(P<0.05). Serum levels of IL-10 significantly increased in OA2h group,compared with other groups(P<0.01). IL-10 levels of myocardial tissue significantly increased in OA6h, compared with control group(P<0.01).The levels of IL-1βgene expression continually increased in myocardium after OA injection(P<0.01). The expression of IL-10 mRNA in the cardium is obviously increased in OA6h(P<0.01).Conclusion: With the changes of serum cardiac enzymes and myocardic pathology in ARDS rat induced by oleic acid, IL-1β, IL-10 protein in serum and myocardial tissue increased, mRNA gene expression enhancing in the myocardium. The changes of IL-1β, IL-10 in the myocardial tissue were possible related to myocardial tissue injury in ARDS rats.
