Aim:With age, muscle atrophy experienced a significant functional decline. While resistance training can increase muscle mass and muscle strength, but such training and the understanding of the mechanism of myogenic response in the elderly animal and human capacity in the muscle hypertrophy are limited, and the effects of changes in training are not clear with age-related cell signaling pathway. Through the comparison of the phosphorylation of mTOR,p70s6k and ERK1 / 2 to observe the effect of aging skeletal muscle protein signaling pathway translation in skeletal muscle about the aged mice after 12 weeks different resistance exercise. In order to explore the mechanism and select effective exercise forms of retard aging, provide the scientific method and the theory of elderly fitness exercise.Materials and methods:This study takes 6-month-old and 15-month-old female Kunming mice as experimental objects. 6-month-old mice for adult control group, 15-month-old mice were selected control group and the elderly exercise group randomly, the elderly exercise group were divided into three groups which named no load swimming group, increasing resistance swimming exercise group and alternate campaign group. In the 24-48 hours after 12-week exercises, take mice bilateral hindlimb shares rectus, weighing, number, add to eppendorf tube, quick-frozen in liquid nitrogen, -80 degrees refrigerator preservation.Application of western blotting molecular biology techniques to get pictures of target protein.Gel image analysis software was used to get the gray value. Gray target protein value divided by internal parameters gray worth relative to the target protein content. Each of the data collected by the average±standard deviation (Mean±SD) said. All the datas are analysis by the statistics software SPSS13.0. The difference between the group were determined by a paired t-test. Difference were considered significant at p <0.05 .Results:1. The weight, muscle mass and muscle protein concentration of aged mice compared with adult mice were reduced significantly;2. The phosphorylation of mTOR,p70s6k and ERK1 / 2 in aged mice compared with adult mice were declined significantly, the capacity of protein synthesis in aged mice were dropped significantly;3. Incremental resistance exercise group and the alternate campaign group could increase the phosphorylation of mTOR and p70s6k in elder mice, but the incremental resistance exercise group increased most significantly, the none load exercise group had no significantly increase;4. Three different resistance exercises can increase the phosphorylation of ERK1 / 2, but incremental resistance exercise group increased the most significantly, followed by alternate exercise group, the none load exercise group increased slightly. Conclusions:1. With the growth of age, the skeletal muscle of elder mice are aging;2. The exercise without load had no significance role in increase skeletal muscle mass and the strength effectively;3. Resistance exercise can activate signaling transduction of protein synthesis in skeletal muscle, enhance muscle protein synthesis, and improve muscle mass and strength, delay the sarcopenia.
