Objective To investigate the proliferation and apoptosis effect of Parthenolide on human non-small cell lung cancer A549 cells in Vitro.Methods MTT assay was carried out to observe the anti-proliferation effect of indicated Parthenolide(0,10,50,100,500uM)on A549 cells in Vitro.The apoptosis of A549 cell induced by Parthenolide was detected by AnnexinV-FIYC/PI double-labeling and low cytometry.The expression ofNF-κB,Caspase-3,Caspase-9,COX-2 was examined by western-blot.Results 1 The inhibitory action of A549 cell growth dealing with parthenolide was observed and showed as a dose-dependent manner.The inhibitor index of different concentration group(10,50, 100,500uM)reached to 8.22±5.02%,16.71±1.66%,21.93±4.59%,33.60±0.65%respectively.And all the groups showed significant difference except the 10uM group.2 The apoptosis index of A549 cell was improved as the dose of parthenolide increasing,as well as the expression of Caspase-3,Caspase-9 accordingly.3 The down-regulation of NF-κB and COX-2 was detected by western-blot assay.The expression diversity between different dozes was also obvious.Conclusion Parthenolide could inhibit the human non-small cell lung cancer A549 cells proliferation in vitro and induce the apoptosis.The apoptosis effect which may relate to the inhibition of NF-κB and the activation of Caspase-3,Caspase-9 is one of the main anti-tumor mechanism of parthenolide.
