Fat particle auto-transplantation means excised and purified fat partic -le being injected into soft tissue defect area for a plump moulding of the recipient site.It is a good kind of filling materials.Because of the high absorption rate,this technique can not be popularized clinically.Many facors influence transplanted fat survival,so many experts did some research works to improve the survival and draw a conclusion:to accelerate angloreestablishment is the key point to improve the transplanted-fat survival. Thus various angio growth factors have become research points.Just like Vascular Endothelial Growth Factor(VEGF).But some researchers think t -hat VEGF can not promote transplanted fat survival directly when it was added to fat particle,.PluronicF-127 is a good kind of material with nontoxic and nonimmunogenic.The present results show that Pluronic micellae may be good microcarriers for drug targeting.until now,there is no report about the effect of PluronicF-127 and VEGF delivery system on grafted fat survival and the restituting blood circulation of pearl fat transplantation.Objective Through establishing a rat fat-transplantation model,injecting PluronicF-127 and VEGF into fat particles,with VEGF function of improving angiogenesis and blood-circulation,to investigate its effect on angiogenesis and survival in recipient area.Method twenty-four female nude mice weighing 20 g were randomly assigned into three groups and treated as follows:we divided fat harvest under same condition into 4 groups,one group as contronl group inflitra-, ded in the another 3 groups of fat respectively in DMEM containing 20ng /ml VEGF,DMEM containing 30%Pluronic F-127,DMEM containing 20 ng/ml VEGF and 30%PluronicF-127,then transplanted 4 groups of fat subcutaneously on the back of groups of mice。after 3 weeks,6 weeks,12 weeks,we dissected fat grafts,measureing their weight retention and put them in histopatholigic examination for HE and CD34.compareing weight retention of each experimental group with that of control and MVD each other.the relative between adipose cell and PluronicF-127 was observed through electron microscope. Results microvessels show brown color after CD34 staining.The MVD and weight of pluronicF-127 and VEGF is significantly more than that of other groups(P<0.05).Conclusion the delivery system of PluronicF-127 and VEGF can significantly improve restituting blood circulation of pearl fat transplantation, and better the survival rate of fat transplantation.PluronicF-127 is a good type of biomaterial,it will be degraded in 6-8 weeks.
