Objective.To sturdy the effect of the NSC transplanted on the MCAO model rats,and to sturdy the relationship between NSC and the two neural nutritional factors:BDNF and VEGF.Methods:80 rats were divided into 4 groups at random:(1)sham operated group,20 rats,only broke the left side external carotid artery;(2)model group,20 rats,only made MCAO model,did not made transplantation;(3)PBS control group,20 rats,transferred 15μL PBS liquid into the rats after 7 days of the operation;(4)NSC transplanted group,transplanted 15μL NSC labeled with Brdu into the rats after 7 days of the operation.The MCAO models were established with suture emboli method.The neurological deficits of rats were evaluated after the transplantation at 24thhour,7thday,14thday 21stday,28thday. Rats cerebral tissue was obtained at 7thday,14thday 21stday,28thday.The distribution of NSC and the change of the BDNF and VEGF factors were investigated by HE dyeing and immunohistochemistry.Results:(1)The neurological deficits of NSC transplanted group was more improved than model group and the PBS control group at 7thday,14thday 21st day,28thday(P<0.05).(2)HE dyeing findings:denaturalization,putrescence and dropsy of nerve cells in NSC transplanted group obviously has improved, compared with model group and the PBS control group;(3) Immunohistochemistry findings:in NSC transplanted group,there were some Brdu positive cells in the infracted hemisphere;(4)There were more BDNF and VEGF factors positive cells in 14thday 21stday at NSC transplanted group than that at the model group and the PBS control group.(P<0.05)Conclusions:(1)The therapy of NSC transplanted into rat brain after artery occlusion can help the recovery of neurological function;(2)The transplanted NSC can enhance the expression of the BDNF and VEGF factors,that may be one protection mechamisms of NSC.
