Part One The establishment and comparison of Rat-Rat heterotopy heart transplantation model in beating heart and arrested heartObjective To establish and evaluate the rat-rat heterotopy heart transplantation model in beating heart with vivo rat replace cardiopulmonary bypass machine-assisted,and rat-rat heterotopic heart transplantation model in modified Heron's methods.Methods We choose 422 healthy,clean-class and male Wistar rats,105 as the donor and 105 as the recipient,and weighing 250~300g,and another 53 for replacing cardiopulmonary bypass machines,159 for blood collection and weighing 280~300g.to establish 53 cases of the rat-rat heterotopy heart transplantation model in beating heart with vivo rat replace cardiopulmonary bypass machineassisted,and 52 cases rat-rat heterotopic heart transplantation model in modified Heron's methods.Results Continuously performed beating heart and arrested heart,the formal experiments test in model of 53 and 52 examples.The model success rate was 94.3%and 96.8%.Conclusion The beating heart transplantation model was a simplified, reasonable,economical,practicable,reliable and high reproducible model without IRI.It was contribute to carry out massive heart beat transplantation in basic studies.Part Two The research of myocardial IRI of heterotopy heart transplantation model in beating heart and arrested heartObjective Establish rat-rat heterotopic heart transplantation model in arrest and beating heart,and evaluate the donor heart edema,peripheral blood myocardial enzyme content,peroxidation products after preservation,compare the difference of beating heart transplantation with the traditional cardioplegia in avoid or mitigate the myocardial Ischemia Reperfusion Injury.Methods To establish rat-rat heterotopic heart trans-plantation model 53 cases in beating heart,and 52 cases rat-rat heterotopic heart transplantation model adopt modified Heron's methods.3 days after transplant,30 recipient are randomly selected from each group and their blood and myocardial tissue of donor heart were collected for determination.Results 3 days after the heart transplantation,detect myocardial water content,myocardial enzymes(cTnI,CK-MB,LDH),malondialdehyde(MDA) of the myocardial tissue,the content of superoxide dismutase(SOD)in serum. The difference of Group A(beating transplant group)and group B(cardioplegia transplant group)groups were significantly(P<0.01).Conclusion In the entire transplantation process of this beating heart transplantation model,the coronary circulation is not interrupted,myocardial be perfused sustained by oxygenated blood and maintain the rhythm of heart beat, provided an environment closest to the physiological state of aerobic metabolism,PH and electrolyte metabolism,which can effectively avoid IRI, and the occurrence of other various shortcomings caused by cardioplegia,access an ideal myocardial protective effects.
