Objective To observe the effects and significance of Herba Erigemotis the expression of vascular endothelial growth factors(VEGF)and pigment epithelium -derived factors(PEDF)in the retinas of diabetic rats.Methods Diabetic rats by streptozotocin(STZ)were randomly divided into diabetes group(10×2)and Herba Erigemotis treatment group(10×2).Another 20 normal rats were recruited as normal control group.The Herba Erigemotis treatment group was injected Herba Erigemotis(20 mg/kg)while diabetes groups and normal control group were injected saline solution into peritoneal cavity once every day.2 weeks and 8 weeks later,10 rats in each group were killed and the expression of VEGF and PEDFin the retinas were detected by RT-PCR and immunohisochemistry.Results:VEGE:In 2 weeks,the expression of VEGF mRNA in diabetic group increased and it were significantly more than that of streptozotocin treatment group(P<0.05),but the level of protein did not have different.In 8 months,the expression of VEGF mRNA and protein in diabetic group further increased and they were remarkably more than that of Herba Erigemotis treatment group(P<0.05).PEDF:In 2 weeks,the expression of PEDF mRNA in diabetic group and treatment group did not have different with the control group(P>0.05).In 8 weeks, the expression of PEDF mRNA in diabetic group further decreased(P<0.05)and the expression of PEDF mRNA of Herba Erigemotis treatment group they were remarkably more than that of the diabetic group(P<0.05).But the expression of PEDF protein did not have any different.(P>0.05)Conclusion Herba Erigemotis injected intraperitoneaUy decreases the expression of VEGF mRNA and the protein;and Herba Erigemotis injected intraperitoneally decrease the expression of PEDF mRNA in retinas of diabetic rats.so that Herba Erigemotis partly suppresses the development of diabetic retinopathy.
