OBJECTIVE To establish the pattern of mouse skin's photo-damage.To observe the effect of gypenosides(GP)on photo-damage and identify the correlation between gypenosides and anti-photo-damage.METHOD We set up twelve groups of Balb/C mouse skin photo-damage's model. We measured the levels of Superoxide dismutase(SOD),Catalase(CAT),Glutathione peroxidase(GSH-Px),glutathione reductase(GR)in the epidermis of mice with visible spectrophotometry and ultraviolet spectrophotometric method.Groups as follow:control group,UVB group,different concentration of gypenosides groups, Vitamine E groups,total 12 groups.RESULTS:(1)The animal models of photodamage were made successfully.The levels of SOD,CAT,GSH-Px,GR in Balb/C Jimpy mice's skin tissue of UVB expose group were lower than those of control group(P<0.05).(2)The levels of SOD,CAT and GR in Balb/C Jimpy mice's skin tissue of 1.5%gypenosides Groupâ… were higher than those of the UVB group(P<0.05).The significant difference of the level of SOD, CAT,GSH-Px,GR was not observed between 1.5%gypenosides Groupâ… and the vitamin El group.(3)The levels of SOD,CAT,GSH-Px and GR in Balb/C Jimpy mice's skin tissue of 1.5%gypenosides Groupâ…¡were higher than that of the UVB group(P<0.05).The significant difference of the level of SOD was not observed between 1.5%gynosaponin Groupâ…¡and the vitamin Eâ…¡group,but the levels of CAT,GSH-Px,GR of 1.5%gypenosides Groupâ…¡were higher than that of vitamin Eâ…¡group.(4)The levels of SOD,CAT,GSH-Px,GR in Balb/C Jimpy mice's skin tissue of 3%and 6%gypenosides Group were lower than that of the control group(P<0.05).CONCLUSION:(1)The levels of SOD,CAT,GHS-Px,GR in the epidemis of mice decreased after UVB irradiation,which were the main enzyme to counteract oxidative damage.(2)The Balb/C mice were topically treated with 1.5%gypenosides,followed by an exposure with UVB,which the levels of SOD,CAT,GSH-Px,GR were advanced.Ruling out the matrix interference,the anti-oxidase effect of 1.5% gypenosides is similar to that of vitamin E.(3)The topical application of 1.5%of gypenosides had the protective effect on UVB mediated skin damage.The protective mechanism may be related to the eliminating the free radicals and increasing the levels of SOD,GSH-Px,CAT,GR to inhibit the oxidative damages caused by UVB. (4)3%and 6%of gypenosides have no anti-oxidative-damage effect.
