Objective Candida albieans insolubleβ-glucan(CAIBG)is a kind of biological reponse modifiers(BRM)which has extensive biological activities,such as anti-tumor activity,increasing the number of leukocytes,anti-virus activity and so on.Previous experiments of our research team isolated successfully CAIBG from the cell walls of Candida albieans,and confirmed that CAIBG was able to elevate the number of leukocyte of peripheral blood in mice and can induce the introduction of TNF-βin blood,and studied the mechanism of them in the molecular level.In this study,insolubleβ-glucan was extracted from Candida albieans,to explore whether CAIBG can suppress the tumor cells growth and vitality in vitro and to search its mechanism.Methods Firstly;insolubleβ-glucan was extracted from Candida albieans,and malignant melanoma B16 cells,gastric carcinoma SCG-7902 cells,hepatoma SMMC cells,pulmonary adenocarcinoma XW-05 cells were cultured in vitro.Secondly,the different concentration of CAIBG was added to cultured tumor cells in vitro,and the morphologic changes of the tumor cells were observed by phase microscope.The reforming MTT assay was used to evaluate the growth inhibition effect of CAIBG on malignant melanoma B 16 cells,gastric carcinoma SCG-7902 cells,hepatoma SMMC cells,pulmonary adenocarcinoma XW-05 cells at different times.Thirdly,the cancer cells' growth inhibitory effects of CAIBG were compared with the Cisplatin. The cell cycle distribution and apoptosis rate of pulmonary adenocarcinoma XW-05 cells were detected by flow cytometry.Results CAIBG had different inhibitory effects to the different tumor cells in vitro,and apoptosis of tumor cells were observed morphologically.The highest growth inhibitory rate of the malignant melanoma,gastric carcinoma,pulmonary adenocarcinoma were 42.8%, 59.88%,61.97%,and IC50were about 1506.5μg/ml,429.8μg/ml,293.4μg/ml respectively,but had no growth inhibition effect to hepatoma SMMC- 7721 cells.The growth inhibitory effect of different concentration of CAIBG to the same cancer cells had variablility(P<0.05).The sensi- tivity of different cancer cells to the same concentration of CAIBG had significant variablility(P<0.05).The pulmonary adenocarcinoma XW-05 cell was the most sensitive one to the CAIBG.The cancer cells' growth inhibitory effects of CAIBG and the Cisplatin had significant variablility(P<0.05).The cell cycle distribution of pulmonary adenocarcinoma XW-05 cells had no marked change after being treated with CAIBG. Conclusions CAIBG had growth inhibitory effect on malignant melanoma B16 cells,gastric carcinoma SCG-7902 cells,pulmonary adenocarcinoma XW-05 cells,and had the most significant effect on pulmonary adenocarcinoma XW-05 cells.Different tumor cell had different reaction to the same concentration of CAIBG,and the same tumor cell had different reaction to different concentration of CAIBG. The mechanism of the tumor cell's growth inhibitory effect of CAIBG in vitro was that probably there were receptors of the CAIBG on the plasma membrane of the tumor cells,therefore influenced the metabolism of the tumor cells which leaded to the cell death.CAIBG is wothy of further research and development.
