| Partâ… The establishment of lung VX2 tumor resection model in rabbits.Objectives:To establish the lung VX2 tumor resection model in rabbits for further investigation.Methods:The VX2 tumor tissues were implanted into the right lung of 12 New Zealand rabbits to form the VX2 transplanted tumor. Under aseptic conditions,the lobe which transplanted the tumor was resected to establish the lung VX2 tumor resection model.Results:The tumorigenic rate was 100%. One rabbit had been rejected because multiple tumors formed in the right lung of the rabbit.The other 11 rabbits received the resection.Only one rabbit died of respiratory failure after the operation,the other 10 rabbits all survived 30 days after the operaion.Conclusions:The lung VX2 tumor resection model in rabbits was easy to establish and the success rate was high,it was believed to be an ideal and reliable animal model for further study.Partâ…¡The exprssed changes of peripheral blood CD4~+CD25~+ regulatory T cells in rabbits with VX2 lung tumors before and after operationObjective:To discuss the influence of the tumors and operation to the immunological function of the rabbits,by means of detection of CD4~+CD25~+T cells in peripheral blood of the rabbits with VX2 lung tumor before and after the operation..Methods:30 New Zealand rabbits with VX2 lung tumors were divided into 3 groups.Group A:the rabbits with lung VX2 tumor received tumorectomy; Group B: the rabbits with lung VX2 tumor received no operation;Group C: the rabbits without lung VX2 tumor received tumorectomy only.The peripheral blood of rabbits were drawn at time before implantion,before the operation,the 7th day after the operation,the 15th day after the operation and the 30th day after the operation.All the samples are analyzed by flow cytometry(FCM). Statistical analysis has been performed with SPSS 15.0,the relationship was examined by t-test or One-Way ANOVA.Results:1 The proportion of the CD4~+CD25~+ regulatory T cells in peripheral blood of the rabbits increased after the implantion of VX2 tumor.2 The proportion of the CD4~+CD25~+ regulatory T cells in peripheral blood of the rabbits with VX2 lung tumor which received tumorectomy obviously decreased at the 15th and 30th day after the operation,but the the proportion of the CD4~+CD25~+ regulatory T cells in peripheral blood of the rabbits with VX2 lung tumor which received no operation did not have the foregoing change.Conclusions:1.The increase of the proportion of CD4~+CD25~+ regulatory T cells in peripheral blood is related to the progression of the tumor.2.The functional disorder of immune system recovers in a certain extent after the resection of the tumor.Partâ…¢The detection and clinical significance of CD4~+CD25~+ regulatory T cells in the peripheral blood of patients with lung cancer before and after operationObjective:To detect the expression level of CD4~+CD25~+ regulatory T cells in the in the peripheral blood of patients with lung cancer before and after operation,and to explore the relative mechanism.Methods: The specimens of 43 lung cancer patients,14 pulmonary benign disease patients and 20 hea1thy adults were collected from July 2007 to Mar 2008 in the Department of Cardiothorcic Surgery of the First Affiliated Hospital of Soochow University. Flow cytometry was used to detect and analyze CD4~+CD25~+ regulatory T cells in the peripheral blood of three groups,meanwhile,it also examine the CD4~+CD25~+ regulatory T cells level at the 7th day 30th day after operation in lung cancer patients.Statistical analysis has been performed with SPSS 15.0,the relationship was examined by t-test or One-Way ANOVA.Results:1The proportion of CD4~+CD25~+ regulatory T cells of lung cancer patients decreased obviously at the 30th day after the operation .2 The proportion of CD4~+CD25~+ regulatory T cells of lung cancer patients was significantly higher than benign group and healthy group .3 No difference was found in the level of CD4~+CD25~+ regulatory T cells of lung cancer patients before resection among different pathological types4 The proportion of CD4~+CD25~+ regulatory T cells ofâ…¢period lung cancer patients was significantly higher than the proportion ofâ… andâ…¡period patients.Conclusions:1 The proportions of CD4~+CD25~+ regulatory T cells increased in the peripheral blood of lung cancer patients which is ralated to the clinical stage suggest that CD4~+CD25~+ regulatory T cells selectively inhibit the immune response and contribute to the progression of lung cancer.2 The proportion of CD4~+CD25~+ regulatory T cells of the lung cancer patients decreased after resection,which suggests indicated that the immune function recovered in a certain extent.3 The proportion of CD4~+CD25~+ regulatory T cells in petipheral blood may become a good cellular immunological index of lung cancer patients in clinica1monitor. |
