Objective: To investigate the expression of E-selectin and CD11b on carotid artery anastomotic stoma in rats and the sffects on earlier period patency rates of the stoma.Methods:A comparative experimental study was performed in 60 Wistar/ST albino rats. One side of carotid artery as a experimental group was end-end anastomosed with suturing,the other side as a control side was only dissected. TA (time for anastomosis) and patency rate were statisticated Milking test and intraoperative microvascular doppler ultrasonography were used to measure patency rate immediately and before harvesting the specimens.By using microvascular doppler ultrasonography tenchnique,the hemodynamic indexes of Vm (mean blood velocity) and RI (resistance index) of anastomosed arteries were measured at the 2-hour, 4-hour,6-hour, 12-hour, 24-hour and 72-hour after operation. The artery specimens were harvested at the same time and were analyzed by light microscopy, immunohistochemistry and RT-PCR. All the experimental datas were statisticated.Results:①T A of the artery anastomosis with suturing was (13.6±1.9)min.The patency rates were 100% immediately after artery anastomosis,and ,however,87% before harvesting the specimens by using Milking test and intraoperative microvascular doppler ultrasonography.There were significant differences of Vm and RI between experimental group and control group at every times stage(P<0.05).With the times lasting after artery anastomosis,the Vm and RI were at the descenting tendency.②The immunohisto- chemistrical results indicated that E-selectin expression was up-regulation on the surface of arterial endomembrane at 2-hour after artery anastomosis ,especially on the parietal thrombus,and that the up-regulation of E-selectin increased most remarkably at 4-hour or 6-hour after artery anastomosis and tapered after 24-hour or 72-hour.However,there was not significant expression up-regulation in the control group because of expression extremely trifling on nomal artery .③Compared withβ-ACTIN ,the RT-PCR results indicated that expression of CD11b was up-regulation in the arterial stoma was up-regulation at 2-hour after artery anastomosis,and increased the most remarkably at 24-hour. There were significant differences between experimental group and control group at every times stage(P<0.05).Conclusions:The patency rates were 100% immediately after artery anastomosis,and , however,87% before harvesting the specimens.The stenosis or occlusion of arterial stoma at earlier period mostly occurred in 24-hour after artery anastomosis.The immuno- histochemistrical and RT-PCR aresults indicated that expression of E-seclectin and CD11b was up-regulation in the arterial stoma was up-regulation .This implied that up-regulation expression of E-seclectin and CD11b would be importantly effective factors on stenosis or occlusion of arterial stoma at earlier period after artery anastomosis.
