The metabolic characters of glucose and fat in gestational insulin resistant rats induced by high fat dietObjeceive:By intervention fat enriched diet,to explore a gestational insulin resistance rats model establishing method,which is steady,reliable and similar to human being.Methods:Nine weeks old female wistar rats adapted to environment for two weeks, after randomly group were assigned to normal diet and high fat diet separately.Exposed to high fat diet for five weeks every group was divided into two subgroups in random,one subgroup of each diet became pregnant.About the eighteenth day of pregnancy administered Insulin Tolerance Test to evaluate the sensitivity of insulin,and on twenty day of pregnancy operated Oral Glucose Tolerance Test.Then executed rats,Weighted visceral fat and liver,sliced and dyed to observe under microscope.Gathering suborbital vein blood to dynamicly test the level of glucose,triglyceride,cholesterol and free fatty acids in blood.Results:After raised high fat diet for two weeks,the weight of rats increased distinctly,and during pregnancy this circumstance continued.Before pregnancy elevated level of glucose,triglyceride,cholesterol and free fatty acids,further elevated in pregnancy time.Areas under curve of OGTT in high fat diet rats were larger than normal diet rats, the same circumstance occurred between pregnant rats and virgin rats.Conclusion:In pregnant rats group exposed to high fat diet visceral fat increased, fatty liver appeared,according to insulin tolerance test insulin resistance was evident. Pregnant rats raised high fat diet exhibited visceral fat gather,metabolism of glucose and fat abnormal,the extent of insulin resistance aggravated.All these traits were consistent with the characters of gestational pathologic insulin resistance in human being.The study on apoptosis of pancreatic beta cells in gestational insulin resistant rats taking high fat dietObjective:The aim of this study is by observing the number change of islets beta cells in gestational insulin resistant rats exposed to high fat diet to explore the influence of lipotoxicity on beta cell apoptosis.Methods:After finishing the first experiment,executed rats,selected pancreatic tail tissue and fixed,further slides were stained with insulin antibody by immunocytochemistry to confirm the location of beta cells.Image analysis system determined mean area stained positive cells in each inslet,which stood for total number of beta cells.The apoptotic beta ceils in islets were detected and quantified by the Tunel technology to calculate apoptosis ratio.Results:Compared mean stained positive area among each group,the largest was gestational rats fed high fat diet,and gestational rats was more than virgin rats,but the difference had no statistic significance.About the total of pancreatic beta cells gestational rats was more than virgin.The frequency of beta cells apoptosis was higher in diet intervened rats,gestational rats was higher than virgin.The same trend happend in the number of positive cells,but discrepancy was not remarkable.Conclusions:After high fat diet feeding,gestational rats pancreatic beta cell apoptosis increased,lipotoxicity may play a certain role in gestational diabetes mellitus occurrence.
