Expression Of RhoGDI Protein In Lung Cancer

Objective:To detect the expression level of RhoGDI protein in small cell lung cancer (SCLC),nonsmall-cell lung cancer(NSCLC)and lung inflammatory pseudotumor tissues,to evaluate clinical significance of RhoGDI expression in lung cancer.Methods:The expression levels of RhoGDI protein in tissues of SCLC(8 cases), NSCLC(include 19 cases of squamous carcinoma and 12 cases of adenocarcinoma)and lung inflammatory pseudotumor(5 cases)were detected by western blotting.The pathologic and clinical data of the patients with lung cancer were also collected.The relationships between RhoGDI expression and the clinical and pathological character were analyzed in order to evaluate the significance of RhoGDI expression in lung cancer.The experimental data were analyzed by SAS 6.12 software.Results:1,The level of RhoGDI protein was significantly higher in SCLC group (0.903±0.228)than that in NSCLC(0.522±0.152)(t= 4.473,p＜0.005)and inflammatory pseudotumor groups(0.485±0.095)(t=4.586,p＜0.005).2,In NSCLC,the level of RhoGDI protein was significantly higher in poorly differentiated tumors(0.649±0.123)than that in moderately and high differentiated tumors(0.458±0.138)(t=3.718,p＜0.05)and inflammatory pseudotumor tissues(t=2.84, p＜0.05).The level of RhoGDI protein did not seem to vary between squamous carcinoma(0.499±0.14)and adenocarcinoma(0.562±0.167)(t=1.082,p＞0.05).3,In NSCLC,the level of RhoGDI protein in tumor tissues was increased gradually when tumor stage of pathology was increase,but significantly difference was not found among them(F=0.6,p＞0.05).The level of RhoGDI protein was not found to related to tumor size(t=1.15,p＞0.05)and lymphatic metastasis(t=1.648,p＞0.05).4,The protein level of RhoGDI in NSCLC was midrange positively correlated to the serum levels of lactate dehydrogenase(LDH)(r=0.46,p＜0.01).5,The protein level of RhoGDI was not related to the hemoglobin,platelet and seralbumin.Conclussion:1,The overexpression of RhoGDI protein in SCLC and poorly differentiated NSCLC indicate that RhoGDI may play a important role in the tumorigenesis, invasion and metastase of lung cancer.2,The level of RhoGDI protein increasing gradually from stageⅠtoⅢ,and a positive correlation between RhoGDI protein level in tumor tissues and serum level of LDH,indicate that there seems to be a relationship between RhoGDI expression and tumor load in NSCLC.
