Objective In the brain, the Na+/H+ exchanger (NHE), a ubiquitous protein, is essential for intracellular pH (pHi) regulation under both physiological and pathological conditions.Recently, it has been reported that Na+/H+ exchanger (NHE) inhibitors demonstrated protective effects on ischemia/reperfusion brain injury in animal models. In the present investigation we evaluated the effects of inhibition of Na+ /H+ exchange with amiloride applied topically onto the rat cerebral cortex prior to ischemia-reperfusion. By contrasting the activity of ATPase,mitochondrial succinic dehydrogenase(SDH) in brain tissue and the number of apoptotic cells in the cortical territory 24 hours after ischemia-reperfusion,the therapeutic effect of amiloride on experimental cerebral ischemia-reperfusion damages can be evaluated and the mechanism of this medicine can be studied.Method 50 adult male Wistar rats were divided randomly into 5 groups. The model group was administrated with isotonic Na chloride.The amiloride group was administrated with different dose of amiloride(1.25mg/kg,2.5mg/kg,5mg/kg). Observing the influence of amiloride on the activity of ATPase,mitochondrial succinic dehydrogenase(SDH) in brain tissue and apoptotic cells in the cortical territory on the acute experimental incomplete cerebral ischemia model induced by ligating bilateral common carotid arteries for 30 minutes.and then reperfusion 24 hours.By constrasting the datas in every group,the effect of amiloride can be evaluated and the mechanism of action can be discussed.Result The activity of ATPase,mitochondrial succinic dehydrogenase(SDH) in brain tissue is much lower in the model group than in the other groups. The activity of ATPase,mitochondrial succinic dehydrogenase(SDH) in brain tissue were a little lower than the sham operated group and higher than the model group.The apoptotic cells in cortex is much higher in the model group than in the other groups. The apoptotic cells in the amiloride groups (1.25mg/kg,2.5mg/kg,5mg/kg) were no more different,but they were lower than the model group.Conclusion Amiloride possessses stronger protection. The present results indicate that amiloride can obviously enhance the activity of ATPase,SDH and renovate the cell membrane and mitochondria membrane and protection the function of mitochondrion.And it has a role of reducing neuron apoptosis after cerebral ischemia-reperfusion.
