Objective: ischemia-like-reperfusion was made on primary cultured rat's neurons.To study the changes of the expression of UCP4 and the level of ROS in primary cultured rat's cortical neurons induced by ischemia-like-reperfusion and the interventional effect of alprostadil.Methods:Cerebral cortical neurons from newborn wistar rats were cultured in vitro.Cortical neurons cultured for 10d were randomly divided into three groups: control group ,ischemia -like- reperfusion group and treatment group.Cortical neurons were exposed to ischemia-like environment for 30min and then cultured for 3h, 6h ,12h under normal condition.Then morphology of nerurons was observed under inverted microscope. Immunocytochemical method was used to investigate the expression of UCP4 at 3h,6h,12h after reperfusion. The flurescent intensity of reactive oxygen species(ROS) was determined by flow cytometry..Results:Compared with control group,the UCP4 expression decreased at 3h,6h,12h after ischemia-like-reperfusion(P<0.05), especially at 6h,12h after reperfusion dramatically(P< 0.01) .while the level of ROS increased gradually( P < 0.05) . UCP4 expression was negatively related to ROS(r=0.745,P<0.05). Compared with ischemia -like- reperfusion group, UCP4 expression increased and the level of ROS decreased at 3h,6h,12h after reperfusion in treatment group(P<0.05).Conclusion:UCP4 may involve in physiological process of ischemia–like–reperfu sion ; and its expression may influence the level of ROS.; Alprostadil may decrease the level of ROS by increasing the expression of UCP4 and was of protective effect on neurons.
