Objective: To study the effect of testosterone propionate on apoptosis and inflammation in brain tissue after focal cerebral ischemia reperfusion in rats.Methods: Sixty healthy male Wister rats were randomly divided into five groups: intact group, orchiectomized group (ORX), ORX treated with testosterone propionate 5mg/kg group (ORXT1), ORX treated with testosterone propionate 25mg/kg group (ORXT2) and ORX treated with testosterone propionate 50mg/kg group (ORXT3), 12 rats in each group . The ORX groups were orchiectomized. After four weeks the ORXT groups were taken different dose of testosterone propionate by hypodermic injection, one time per day。The middle cerebral artery occlusion (MCAO) model was made by the suture method after 7 days . After middle cerebral artery occlusion for 2 hours and reperfusion 24 hours, the infarct volume and the neurologic deficit were tested ;the levels of Bcl-2, Bax and TNF-αwere detectived by immunohistochemistry technology.Results:Neurologic impairment in various degree appeared in the contrast group rat after CI/RP.The infarct area, where showed white colour because not stained by TTC, can be distinguished clearly to the normal brain where showed red colour. Propein Bcl-2, Bax and TNF-αcan be etectived by immunohistochemistry technology.1. In the intact group,scores of neurological deficit and infarct volume were uniformity compared with the ORX group(P>0.05).2. In the low dose of testosterone propionate preconditioning groups, scores of neurological deficit and infarct volume were high compared with the ORX group and show statistical differences (P<0.05) .3. In the middle dose and the high dose of testosterone propionate preconditioning groups, scores of neurological deficit and infarct volume were high compared with the ORX group and show statistical differences significantly ( P<0.01).4. In the intact group,levels of protein Bcl-2, Bax and TNF-αwere uniformity compared with the ORX group(P>0.05). 5. In the low dose of testosterone propionate preconditioning groups, levels of protein Bcl-2, Bax and TNF-αshow differences compared with the ORX group and show statistical differences (P<0.05) .6. In the middle dose and the high dose of testosterone propionate preconditioning groups, levels of protein Bcl-2, Bax and TNF-αshow differences compared with the ORX group and show statistical differences significantly (P<0.01) .Conclusion:1 Testosterone propionate preconditioning can increase the neurologic deficit degree after the focal cerebral ischemia reperfusion.2 Testosterone propionate preconditioning can maxify the infarct volume after the focal cerebral ischemia reperfusion.3 Testosterone propionate preconditioning can aggravate the neuro apoptosis of ischemic area via enhancing the expression of protein Bax and decreasing the expression of protein Bcl-2.4 Testosterone propionate preconditioning can aggravate the inflammation in the process of the focal cerebral ischemia reperfusion, and its mechanism is related to enhancing the expression of protein TNF-α.
