Object Poly(L-lactide) and the pig acellular cartilaginous matrix were mixed and gathered to prepare one kind of new scaffold,PLLA/acellular cartilaginous scaffold,and each scaffolds were analysed of their characters,pore sizes,biological mechanic characters,water-binding capacity, biological degradability and so on.Then they were discussed if one of them is an ideal new scaffold for cartilage tissue engineering.Methods A three-dimensional polymer scaffold was developed by mixing PLLA with acellular cartilaginous matrix using a thermally induced phase separation technique(TIPS) under the different proportion of mixture,the different temperature.The average pore sizes of different scaffolds under different temperature in each thin slice were analyzed with an image analyzer. The porosity of the polymer scaffold was determined by the drainage.Their elasticity coefficients were determinated under the room temperature using the biological mechanics determinant.Water-binding capacity and biology degradability were analysed with the formula of(wet weight-dry weight)/the dry weight and d=(W0-Wt)/W0×100%.The statistics processing uses SPSS13.0 to carry on the analysis.The analyzes make use of variance analysis and the LSD law.Resuls The statistic results of different temperatures of the polymer scaffold pore sizes,porosity, the water-binding capacity,the biological mechanics character,the biology degradability under the different proportion of mixtures have been shown.Regarding the support aperture,the compositions of different materials were not the main reason but the different temperature was the key step to it.Each group were compared with no difference statistically(p<0.05) under the different temperatures.Regarding the factor of porosity and water-binding capacity,each groups of the different temperatures among the different materials were compared with no difference statistically,but the different composition of scaffolds material were with obvious difference. Regarding each kind of scaffolds material prepared under the different temperature,the biological mechanic characters of each kind of scaffolds were with no significance difference. The statistic analysis of the biological degradability under different proportions of mixture and the different temperature polymer scaffolds demonstrate a significance difference.The PLLA/acellular cartilaginous sandwich composite was made,mimicking the natural cartilage both in composition and in microstructure.Its various characteristics have been basically conformed to the requirement of a cartilage scaffold.Conclusion In brief,A three-dimensional polymer scaffold was developed by mixing PLLA with acellular cartilaginous matrix using a thermally induced phase separation technique(TIPS) under the different proportion of mixture and the different temperature.Each kind of scaffolds pore size,the biological mechanics character,the water-binding capacity,the biology degradability and so on were analysed with suitable methods.The pore size at situated temperature will be between 100um~300um,the shape is clear naturally with an orientation rule, the acellular cartilaginous matrix was distributed evenly in the three dimensional structure constructed by poly(L-lactide),and the three dimensional porous scaffold was with a satisfying overall shape and microscopic structure.It was shown the new scaffolds conform to the requirements of a tissue engineering scaffold under situated temperature and proportion of mixture at the aspects of pore size,the biological mechanic character,the water-binding capacity, the biology degradability and so on.The novel three-dimensional PLLA/acellular cartilaginous matrix scaffold is likely to be applied in the cartilage tissue engineering.
