Purpose:To explore mechanism of action on apoptosis of Parkinon disease rat models with electroacupuncture scalp point-through-point therapy.Approach:The Parkinson disease rats were modeled by injection in the left striatum with 6-hydroxydopa mine and treated by the elect- roacupuncture scalp point-through-point therapy.All the experiment- tal rats were divided into four groups:Nor ma l Group,Sha m- modeling Groups,Model Group and Acupuncture Group.The Acupuncture Group were treated with the electroacupuncture scalp point-through-point therapy and compared to the Shan-modeling groups .(1)He ma toxylin and eosin stain methods are adopted to obse rve morphologic change of substa ntia nigra;(2)The method of i mmunohistochemistry is adopt- ed to detect positive prosop-density and integrating photodensity of tyrosine hydroxylase(TH) and brain-derived neurotrophic factor (BDNF) in substantia nigra;(3)The method of Ter minal deoxynucle- otidyl transferase-mediated dutp nick-end-labeling(TUNEL) is adop- ted to detect the apoptotic quantity in the substantia nigra;(4)The method of high pe rfor manc e liquid chromatogram( HPLC)- ultraviolet is adopted to dete ct content of hydr oxytyramine( DA)in the striatum.Results: (1)Compare with the model group,the nerve cell number of substa ntia nigra in the destructive side of the acupuncture group is obviously increased,cell atrophie and de generation is obviously relieved;(2)Compared with model group, the proteic expression of TH and BDNF of acupuncture group is obviously increased(P<0.05)and decreasded of nor ma l group and sha m- modeling group (P<0.01);(3)Compared with model group,the quantity of apoptosis of acupuncture group is obviously decreased(P<0.05) and increased of nor mal group and sha m- modeling group(P<0.01);(4) Compar ed with model gr oup,the DA content of striatum of acupuncture group is obviously increased(P<0.05),and decreased of normal group and sham-modeling group (P<0.01).Conclusion: The electroacupuncture scalp point-through-point therapy can impr ove circling behavior of PD mode l rats,change histomor phology and enha nce TH and BDNF proteinum expres sion of substantia nigra of PD model rats,and decrease apoptotic quanti- ty,and increase DA cotent.It hints that the electroacupuncture scalp point-through-point therapy enhance pos sibly TH and BDNF activity,so striatum dopa mine contents increased and apoptosis amount decrease.
