ObjectiveTo study the therapy effect and possible mechanism of hepatic stimulator substance (HSS) associated Runganling on nonalcoholic fatty liver disease (NAFLD) of rat.Methods1.HSS was purified from porket livers.The purification steps included ultrasonication,heat denaturation, alcohol precipitation, et al.The growth effects of HSS on SMMC-7721 cells were measured by [3H]-TdR incorporation method.2.A kind of Chinese traditional medicine preparation named Runganling was developed,which mainly consists of Underleaf pearl,Black bean,Salvia miltiorrhiza,etc.3.NAFLD model was induced in SD male rats by feeding with a diet rich in lipids for 8 weeks.After treating with HSS,Runganling and their combination expectively,the pathology and type-B ultrasonic of the liver were observed,the serum biochemical and liver TG parameters,oxidative/anti-oxidative parameters and early fibrosis parameters of liver tissue were measured.Results1.HSS significantly promoted the incorporation of [3H]-TdR into DNA of SMMC-7721cells.2.Observing the pathological changes of liver under light lens,we received the result that group treated with HSS or Runganling had definite effect in improving hepatic steatosis and HSS unite Runganling treated group had the best effect comparing with model control group.3.Observing the image of liver under type-B ultrasonic,we concluded that group treated with HSS or Runganling had evident effect in weakdening the echo of liver parenchyma and HSS unite Runganling treated group was similar to the normal.4.The elevation of TG content of hepatocytes and ALT, AST, TG level of blood serum induced by NAFLD were restored remarkably by HSS or Runganling,especially their combination and there was dignificant difference compared with model control group.5.The elevation of MDA content of liver tissue and the less of SOD activities induced by NAFLD were restored remarkably by HSS,Runganling and their combination,and the difference between HSS unite Runganling treated group and model control group was statistically distinctive.6.Both elevation of HYP content and expression of TGF-β1, PDGF-B of liver tissue were restored by HSS,Runganling and their combination,and the difference between HSS unite Runganling treated group and model control group was statistically distinctive.Conclusion1.HSS purified by this method can effectively stimulate the growth of hepatocytes.2.Both HSS and Runganling can restore NAFLD-induced liver damage, the effect of the former maybe due to anti-oxidative activity and improving the disorder of fatty metabolism and the latter maybe due to improving microcirculation and inhibiting fibrosis of liver.Therefore,combination of two drugs can enhance curative effect.
