| ObjectiveTo observe the effects of compound DANSHEN dripping pill (DSP) on artery elastic function and the effects of compound DANSHEN dripping pill (DSP) on the relaxation tension of excised thoracic aorta annulation in rats and to investigate its related mechanisms.Methods1. 76 cases of arterial stiffness patients and 35 healthy volunteers were entrolled in this study. after 30 minutes, brachial-angle pulse wave velocity (baPWV), large artery elasticity index (C1) and small artery elasticity index (C2) were measured in 35 arterial stiffness patients with 10 DSP pills, 41 arterial stiffness patients with 20 DSP pills and 35 healthy volunteers with 20 DSP pills sublingually .2. Isometric tension was recorded in response to drugs in organ bath. Effects of DSP(2.7~21.6 g/L) on the vascular tone of resting,high potassium chloride (KC1) and norepinephrine (NE) pre-constricted rat thoracic aorta with or without endothelium were determined.Results1. A significant reduction of the baPWV (P<0.05), increasing of the C1(P<0.05)and C2(P<0.05)were observed in the arterial stiffness patients with 20 DSP pills and the healthy volunteers. Only small artery elasticity index(C2) (P>0.05)were increased in the arterial stiffness patients with 10 DSP pills.2. DSP(10.8~21.6 g/L) caused concentration-dependent relaxation of the aorta rings pre-constricted with NE in the endothelium-intact rings. DSP(21.6 g/L) relaxed the aorta rings without endothelium in the pre-constricted rat thoracic aorta with NE, DSP (5.4~21.6 g/L)obviously vasodilated the rat aortic rings pre-constricted by KCl with or without endothelium,which was concentration-dependent.Conclusion1. DSP improved artery elastic function immediately.2. The relaxation of DSP in the rat thoracic aorta ring may mediate in both endothelium dependent and endothelium independent manners.3. DSP can improve artery elastic function through vasodilatation of vessel. |
