Objective:To detect the expression and distribution of aquaporinl (AQP1)in adenocarcinoma of the lung and to analyze the correlation between AQP1 expression and clinicopathological characteristics and also to provide new potential targets for therapy of lung adenocarcinoma.Methods:1,The lung tissue of 30 cases of pulmonary adenocarcinoma and 30 normal tissues beside tumors were sampled immediately after surgery from the First Affiliated Hospital of Nanjing Medical University(From Mar.2006 through May.2007). The expression and distribution of aquaporinl in adenocarcinoma of the lung was detected by immunohistochemical and western-blot analysis.2,Statistics analysis:application of SPSS 13.0.Results:1,The Sporadic expression of AQP1 was found in the vascular endothelial cells of normal lung tissues.The majority of AQP1 expression could be detected in the vascular endothelial cells, tumor cells in adenocarcinoma of the lung;The positive index of AQP1 protein in tumor tissues(0.8460±0.0782)was remarkably higher than that in normal tissues beside tumors(0.4202±0.0539)(P<0.01);2,The expression of AQP1 protein was detected in both tumor tissues and normal tissues beside tumors,and it was significantly higher(1.3254±0.1785)than that in normal tissues beside tumors(0.0960±0.0212)(P<0.01);3,The expression of AQP1 protein in adenocarcinoma of the lung was associated with both differentiation degree and lymph node metastasis(P<0.05):Expression of AQP1 protein was negatively related with differentiation of lung adenocarcinoma,and positively related with its lymph node metastasis,but it was no correlation with age,sex,TNM stages (P>0.05). Conclusion:The expression of AQP1 was increased in adenocarcinoma of the lung over normal lung tissues.The up-regulation of AQP1 expression in adenocarcinoma of the lung was correlated with both differentiation grade and lymph node metastasis.It indicates that AQP1 might be participate the genesis and development of adenocarcinoma of the lung and it may be identified as targets for therapy of lung adenocarcinoma.
