| Objective To study the role of UⅡin human renal fibrosis.Methods Immunohistochemistry was exployed to determine the expressions of UⅡ,α-SMA,FN and COL-Ⅲin the renal tissue of 32 patients with MsPGN and 6 nomal controls;NRK52E were cultured in rUⅡ(0,10-8mol/l,10-7mol/l),their proliferations were measured using MTT assay,the expressions ofα-SMA,FN and COL-Ⅲwere examined by immunohistochemistry stain.Results In nomal and MsPGN renal tissue,UⅡwas mainly expressed in renal tubular epithelial cells,the intensity of UⅡexpression was significantly increased in moderate and severe MsPGN thanthe control(P<0.05);the intensity of UⅡexpression was positively correlated with the degree of glomerulonephritis (rs=0.324,P=0.047);the expression ofα-SMA,COL-Ⅲin renal tubular epithelial cells with MsPGN was positively correlated with the degree of glomerulonephritis and UⅡexpression(P<0.05);NRK52E were cultured in rUⅡ(0,10-8mol/l,10-7mol/l), after 48 hours rUⅡreavels a mitogenic effect on NRK52E,after 72 hours the expression amounts ofα-SMA,FN and COL-Ⅲwere enhanced(P<0.05).Conclusion UⅡmay play an important role in development of renal fibrosis associated with phenotypic change and increased ECM production and deposite. |
