AimIschemic Cerebrovascular Disease is the common disease,frequently encountered disease in nervous system,because its'rates of death and mutilation are very high,people's life security and quality were influenced severely.After cerebral artery occlusion,if cerebrovascula blood supply be rebounded in effective time limit, the injure of ischemic cerebrovascula will be recoverd.In recent years investigate discovered that after ischemic cells'injury,heat-shock protein will be expressed and will protect the injured cells.Heat-shock protein 70 conducted as molecular chaperones,participate brain tissue's nervous protection after ischemic.Heat-shock protein 70 is the sensitive and reliable molecular biology sign when brain cell or brain tissue encountered reciprocity damage.Whether can elevate therapy cerebral of effectcerebral ischemia by strengthening endogenous injure of brain tissue already becomed the new studied field in international.NuoDiKang Capsule is purifid from bethel integripetal rhodiola herb which is a medicinal plant produced from Tibet plateau,it'sessential components are rhodioside,flavone,integripetal rhodiola herb amoxcillin etc.NuoDiKang Capsule possess the roles of depressing blood viscosity,minicirculation amelioration and two-ways regulation transmit of neurohumor.This empirical study have observed NuoDiKang Capsule's action to rats'cerebral infarction volume,nervous function,pathobiology changes,HSP70 expression in nerve cells after the focal ischemical reperfusion injury,to intend approach NuoDiKang Capsule treat cerebral arterial thrombosis patients's mechanism of action,and to offer dependable evidence clinically to use NuoDiKang Capsule's dosage correctly.MethodsSixty healthy male Wistar rats,supplied from animal experiment center of Lanzhou university,were randomly divided into sham operation(Sham)group, ischemia/reperfusion(I/ R)group,NuoDiKang Capsule high dosage group,NuoDiKang Capsule low dosage group(There were fifteen rats in every group). NuoDiKang Capsule groups were given NuoDiKang Capsule at doses of 0.672g/kg and 0.336g/kg.Sham and I/R group was given NS at dose of 8ml(about 10 mg/kg).The drugs were given twice a day and consecutively for four weeks before operation.The rats were chosen to establish focal cerebral ischemia reperfusion models with middle cerebral artery thread embolism method.after animal model were successfully established,neurological deficits were measured,the scores are all in 2-3,assuming to enter the groups after the rats were awake by means of levo circling when crawlling,left anterior limb adducting and flexion when lifting tails,besides subarachnoid hemorrhage.After the operation,the rats were back to cages,eating food and water freely.Detecting contents consists of four aspects:1.give a mark to rats of neurologic impairment:each group be undertook after 2h ischemia,6h reperfusion injury;to observe NuoDiKang Capsule's action to rats' nervous function after cerebral ischemical reperfusion injury.2.measure rats'cerebral infarction volume:brain tissues of rats oin each group be TTC dyeing after 24h ischemia reperfusion injury,to observe NuoDiKang Capsule's action to rats'cerebral infarction volume.3.observe NuoDiKang Capsule's action to rats'nerve cells after cerebral ischemical reperfusion injury:brain tissues of rats in each group be HE dyeing after 48h ischemia reperfusion injury,to observe pathomorphology changes and calculate density of abnormal nerve cells in cerebral cortex and hippocamp.4.observe NuoDiKang Capsule's action to rats' HSP70 expression in nerve cells in each group:brain tissues of rats oin each group be immunohistochemistry dyeing after 48h ischemia reperfusion injury,to observe HSP70 expression in rats'brain tissue.ResultsHigh dosage NuoDiKang Capsule could obviously increase the HSP70 expression in cerebral cortex and hippocampus,compared with I/R group,NuoDiKang Capsule low dosage group,difference have significance(P<0.05);High dosage NuoDiKang Capsule could improve rats' nervous functional disturbance,minificate rats'cerebral infarction volume,ameliorat degeneration,necrosis ect.patho- damage of brain tissue after focal cerebral ischemical reperfusion injury.ConclusionsNuoDiKang Capsule can relieve ischemia reperfusion injury of brain tissue and improve nerve function lose.The mechanism may be related with the increasing of HSP70 expression,thus increase brain cell's tolerance to ischemic,oxygen deficiency, and create immunological regulation,rejecting injure of free radical,rejecting apoptosis of brain cell after oxygen deficiency,resisting lethal damage,protecting brain cell。...
