Objective: To explore the expressions of heat shock protein 22(Hsp22) and c-fos,and the protecion of minocycline through the expressions of Hsp22 and c-fos in gerbil brain tissue following transient cerebral ischemic reperfusion.Methods:(1).Animal Groups:Fifty healthy male Mongolian gerbils,devided into fourgroups:Normal Group (n=5); Sham Operation Group (n=5); Ischemia Reperfusion Group (I/R) (n=20); Minocycline Intervention Group (MI/R) (n=20); I/R Group and MI/R Group are devided into four subgroups (each group n=5): 6 hours, 1 day , 3 days ,7 days after I/R ingury;(2).Models: 10 minutes'comon carotid artery occlusion(CAO) of both sides and reperfusion for 6 hours, 1 day , 3 days ,7 days in Mongolian gerbils; The Sham Operation Group only give segregation but no occlusion; peritoneal injection of Minocycline (45mg/ kg. Bid) is given in the MI/R Group;(3).Histological analysis of tissue injury: staining with stained with haematoxylin and eosin haematoxylin and eosin (HE).(4).Immunohistochemical Methods.(5).Experimental results were analyzed with the statistic soft package of Spss13.0Result:(1).Staning with HE: The tissues of normal group and sham-operated group was integrate ,but that of I/R were changed with gliocyte hyperplasia and hypertrophia, interstitial edema, cellular edema, gliocyte and neuron edema, neuron necrosis.(2).Immunohistochemistry: There were a few expressions Hsp22 and c-fos in both normal control group of 5 gerbils and sham operation group of 5 gerbils, and there was no significant difference between the two groups (P>0.05). Compared with normal control group, both of the levels of c-fos and Hsp22 in neurons and gliacytes of ischemia-reperfusion gerbils were evidently upregulated at the 6th hour, 1st day, 3rd day and 7th day,and peaked at the 1st day (p < 0.05) and the 3rd day (p < 0.05), respectively, then both downregulated gradually. Compared with I/R group, levels of c-fos in neurons and gliacytes of gerbils in minocycline intervention group were evidently decreased at the 6th hour, 1st day, 3rd day and 7th day (p < 0.05), levels ofHsp20 upregulated, inversely.Conclusions:(1).Expressions of Hsp22 and c-fos in neurons and gliacytes of gerbils weresignificantly increased following cerebral ischemia-reperfusion,this suggested that the increased expressions of Hsp22 could have protected the brain tissue from damage.(2).A correlated relationship was observed between Hsp22 and c-fos during brain I/R injury.(3).Injury of brain tissue after I/R in minocycline groups became moderate. In addition, minocycline may down-regulated levels of c-fos and upregulated levels ofHsp22.
