| ObjectiveTo investigate the effect of Triptolide(TP)on the cell growth and the expressions of mTOR,Survivin,COX-2 mRNA in human Malignant Melanoma A375 Cells to further explore the Anti-tumor mechamisms of TP.Methods1.Human Malignant Melanoma A375 Cells were cultured in vitro and treated with different concentrations of TP.Inverted microcopic, Methylthiazolyldiphenyl-tetrazolium(MTT)assay and Flow cytometry were used to measure the changes of cell morphology,cell proliferation, apoptotic rate and the phase of cell cycle;2.Semi-Quantitative Reverse Transcription Polymerase Chain Reaction(RT-PCR) was used to detect the expressions of mTOR,Survivin, COX-2 mRNA after the treatment of different concontrations of TP in human Malignant Melanoma A375 Cells.Results1.TP revealed a marked antiproliferative effect on the growth of A375 cells in a time-and dose-dependent manner;FCM analysis indicated TP can induce apoptosis of A375 cells and block cells in G2/S phase; 2.mTOR,Survivin,COX-2 mRNA were significantly expressed in untreated A375 Cells and TP treatment could downregulate their expressions,a significant decrease was observed after treatment of 5 and 10ng/ml TP compared with the untreated goup(p<0.05).ConclusionThese results demonstrated that TP could exert anti-tumor effect by effectively inhibiting proliferation,inducing cell apoptosis and downregulating the expressions of mTOR,Survivin,COX-2 gene mRNA. in A375 cells. |
