Heat shock factor1 is one of heat shock transcription factor which is found earliest. In every stress reaction especially in heat shock response it play an important role by regulation HSPs'express.Objective:We set up cell empyrosis model by using empyrosis blood serum to stimulate RAW264.7 cell line, and exosomatic research HSF1's anti inflammatory effect.Methods:We detect HSF1 and NF-κB's expressing by using Western blot and the interaction between HSF1 and HMGB1 \IL-10 by using Electrophoretic Mobility Shift Assay(EMSA).Results: (1)we have found that both HSF1 and NF-κB are obvious increasing in normal team than in control team. In normal team can nearly find HSF1.(2) the result of EMSA has shown HSF1 do have effect with promoter of HMGB1 and IL-10. (3)super EMSA can not detect NF-κB have interaction with HSF1 and HSE which has effect with express controlling of HSF1 and HMGB1.Conclusion: (1) After stimulate empyrosis blood-serum RAW264.7 cell can express HSF1 which can restrain Inflammatory factors' expression to protect itself ,also demonstrate empyrosis model has been established successfully. At the same time the increase of NF-κB shown that it can induce quick inflammatory. (2) In advanced stage of inflammation HSF1 started to express and combine with promoter region of HMGB1 and IL-10 which can restrain HMGB1's express and enhance IL-10's express to feedback regulate Inflammatory reaction.(3) In super EMSA we can not find interaction between NF-κB and HSF1-HSE so we conclude NF-κB has no direct effect with HMGB1 and IL-10 in inflammatory.
