Objective:To compare the protective effects of endotoxin preconditioning and ischemic preconditioning to the warm ischemia reperfusion injury of the rat liver.Methods:The local 70% liver ischemia-reperfusion injury of rat model was used, 36 rats were randomly divided into 3 groups: (1) IR Group ( N=12): animals were subjected to 30min liver ischemia followed by 1 hour of reperfusion ; (2) IP Group ( N=12): before ischemia-reperfusion, the ischemic preconditioning was achieved with 5mn of local ischemia stimulus followed by 5min of reperfusion twice; (3) EP Group ( N=12): before ischemia-reperfusion, the endotoxin preconditioning was achieved with intraperitoneal administration of lipopolysaccharide (LPS) 0.25mg/kg every 24 hours twice. The sham operated group was set for negative control separately. Every animal was sacrificed to take samples when the liver was reperfused for 1 hour, the concentration of ALT,AST in the peripheral blood, the TNF-αand MDA levels in the liver tissue homogenate and the histopathological changes of the liver were examined.Results:(1) The concentration of ALT,AST in the peripheral blood in the IP and EP group were significantly lower than the IR Group (p <0.05) ,and the pathological examinations also showed that the liver injury in IR Group was more severe than in IP and EP Group .(2) The liver tissue homogenate TNF-αand MDA levels were significantly lower in the IP and EP Groups in comparison with the IR group . (p <0.05)(3) The measured values of every index between Group IPand EP has no significant statistical different (p >0.05) .Conclusion:(1) Both EP and IP have protective effects in the liver ischemia-reperfusion injury.(2) There are some relationships between the EP and IP mechanisms of protection(3) The protective effects of EP and IP were nearly equal in our experiment.
