OBJECTIVE:To explore the relationship between insulin-like growth factor-Ⅰ(IGF-Ⅰ),insulin-like growth factor-Ⅱ(IGF-Ⅱ)and fetal growth restriction(FGR).Moreover the connection activity of superoxide dismutase(SOD)and glutathione peroxidase(GSH-Px)and the concentration of lipid peroxide(LPO)and nitric oxide(NO)with fetal growth restriction was also investigated.METHODS:Choose clinic and hospitalizing mid-pregnant women accompanimented with FGR(n=23)in affiliated hospital of central south university from June 2005 to June 2006 as experimental group,artificial labor synchronization mid-pregnant women(n=16)as the control group. The gestational period is 15~27 weeks.Choice pregnant women accompanimented with FGR(n=70)with gestational period of 28~37 weeks including 25 clinical patients and 45 hospitalization ones and normal pregnant women(n=70)with same gestational period.Select 50 delivery women with or without FGR respectively with gestational period of 38~42 weeks.The age of all pregnant women is from 24 to 30.Draw amniotic fluid with amniocentesis,collect venous blood of pregnant women with aseptic technique and cord blood while childbearing. Determine the concentration of IGF-Ⅰwith immunoradiometric assay (IRMA)and that of IGF-Ⅱwith radioimmunoassay(RIA).The method of pyrogallic acid autoxidation and DTNB direct colouration was used to assay SOD and GSH-Px activity respectively.The method of improvement thiobarbituric acid and nitrate reductase was used to assay the concentration of LPO and NO respectivly.RESULTS:(1)The concentration of IGF-Ⅰis reduced significantly in maternal blood and amniotic fluid with mid-pregnant(P<0.01).That of IGF-Ⅱin amniotic fluid is reduced significantly(P<0.01).But the concentration of IGF-Ⅱin maternal blood isn't obviously alteration(P>0.05).(2)The concentration of IGF-Ⅰand IGF-Ⅱin maternal blood,amniotic fluid and cord blood is reduced significantly respectively in Later-pregnant(P<0.01).(3)The concentration of IGF-Ⅰis reduced significantly in maternal blood while delivery(P<0.01),but that of IGF-Ⅱin maternal blood isn't obviously alteration(P>0.05).The concentration of IGF-Ⅰand IGF-Ⅱin amniotic fluid and cord blood is reduced significantly respectively while delivery(P<0.01).(4)The activity of SOD and GSH-Px is reduced significantly in amniotic fluid (P<0.01),but the concentration of LPO is increased significantly in amniotic fluid(P<0.01).(5)The concentration of NO is increased significantly in maternal blood(P<0.01)and cord blood(P<0.001) respectively.The concentration of NO in cord blood is significantly less than that in maternal blood in two groups(P<0.05).The concentration of LPO and NO is positive correlation in maternal and cord blood in two groups.Coefficient correlation is 0.8369 and 0.7763 in maternal and cord blood respectively in experimental group,that is 0.5487 and 0.6217 respectively in control group.Every coefficient correlation is significant (P<0.01).CONCLUSION:1.These reseach incidate that the concentration of IGF-Ⅰis decreased significantly in maternal and cord blood and amniotic fluid.The concentration of IGF-Ⅱis decreased significantly in cord blood and amniotic fluid,but is not obviously altered in maternal blood.The decrease of level of IGF may be one of causes to FGR.To detect the concentration of IGF-Ⅰand IGF-Ⅱis one of indexs of early diagnosis FGR.2.Oxygen free radical(OFR)correlates with FGR;LPO and NO participate development of FGR and they are synergistic.Detecting activity of SOD and GSH-Px and concentration of LPO and NO conduce to diagonosis and prophylaxis FGR.
