This model of diabetes induced by high lipid and glucose diet plus injection of low does Streptozotocin displayed high serum glucose, high serum insulin and IR, which was similar with human. Pathologically, the islet struct display atrophy and destruction, the quantity of cell also decreased. Our further experiment observe the changes of pancreaticβ-cell KATP channels in diabetes rats, so as to study on the influence of nateglinide and tanshinol to the KATP channels, the therapeutical effect and regulating mechanism of these two medicines on OGTT, fasting blood glucose (FBG) and insulin level of DM rats. Model rats displayed impaired glucose tolerance (IGT), FBG markedly increased (p<0.05) and insulin in serum markedly decreased (p<0.05). Nateglinide and tanshinol may effectively improve those abnormalities (p<0.05). Nateglinide displayed a better therapeutic efficacy. Pathologically, destruction of pancreatic small ducts and acini appeared, with some clearly necrosis and congestion in vascellum. Inflammatory cells most of which were lymphocytes infiltrated with halo around them. Structure of pancreatic islets was destroyed with decrease ofαandβcells. Some tissue demonstrated fiber proliferation. Reparation of destructive pancreatic islets was observed in Western medicine group with increasedαandβcells comparing with model group. Decreased number of inflammatory cells and lessening fiber proliferation was observed in Chinese medicine group. Western blot showed that expression of protein Kir6.2 and SUR1 in model rats significantly diminished comparing with what in control rats (p<0.05). Nateglinide displayed up-regulation to the expression in some degree. Tanshinol almost had no influence to expression ofβ-cell KATP channels. These results agreed with those showed from immunofluorescence. RT-(nested)-PCR showed that Kir6.2 and SUR1 mRNA of model rats significantly diminished compared with what of control rats (p<0.05). Nateglinide displayed up-regulation to the expression in some degree, but the expression was still notably lower than control. Tanshinol almost had no influence to expression of and SUR1 mRNA. The model of diabetes induced by Streptozotocin demonstrated IGT, abnormality of FBG and insulin level in serum. Model pancreas displayed extensive acinar degeneration and overt damage of islet, accompanying with chronic inflammation and fibrosis of tissue. Protein and mRNA of KATP channels on pancreaticβ-cells diminished in model, and improvemet happened in various degree and various point of view after treated with nateglinide and tanshinol. Treatment of integrated Western and Chinese medicine will showed better curative effect and prospect.
