ObjectivePaclitaxel(trade name Taxol)is a naturally diterpenoid found in the Taxus, which has great potential as anti-cancer drugs.Paclitaxel does not dissolve in water. Now Paclitaxel injection uses carrying agent of cremophor and absolute alcohol (50:50)which assist to dissolve.Cremophor has affect which promote histamine release and often cause severe hypersensitiveness reaction.Nanometer class polymer particle,which is used as carrying agent of delivering and controlled releasing drug,is one kind of new controlled release system drug.It can penetrate tissue space and be absorbed by cell.It can pass human body minimal blood capillary.In the nanometer controlled release system,releasing mechanism of drug may be drug come out by capsule wall leaching,leaking,diffusing and by corrosion of base material.The keynote of research is that utilizing paclitaxel ordinary preparation prepares paclitaxel deuto-microballoons dosage form,which boosts its bioavailability of anti-mouse breast cancer,enhances drug action,degrades adverse reaction.To search the antitumor and induced apoptosis effect of Paclitaxel-NP on BCML-299-bearing mice in vivo.Methods The tumor inhibition effect was observed in BCML-29-bearing mice. The growth inhibition effect,optical microscopy,the expressein of bax mRNA by situ hybridization and labeling,flow cytometry were were used to examine the effect of Paclitaxel-NP on BCML-299 breast cancer.Results Tumor ininbition was observed in BCML-299 following a 10-day treatment with 10,15mg/kg of Paclitaxel-NP,with tomtal growth inhibition of 76.67%,64.74%,and showed significant change compared with taxol(P<0.0.05).The expression of Bax mRNA was observed in three Paclitaxel-NP group in a concentration-dependent manner,showed significant change(P<0.0.05)between each other.The expression of Bax of the 15mg/kg and 5mg/kg Paclitaxel-NP group showed significant change compared with taxol group(P<0.0.05).The expression of aproapoptotic Bax was increase.The cell showed G2/M arrest and apoptosis.Conclusion Paclitaxel-NP may significantly inhibit the groth of bread cancer in mice by inducing apoptosis and inhibiting proliferation of tumor cells.
