Objective: The MFH is one of the most common malignant tumors in our country, which is characterized By a low respectability rate and a low long-term survival rate. The 1-year survival rate of MFH is under 90%, and the 3-year survival rate is only 52%, thus the prognosis is one of the worst among the STS. Invasion and metastasis is considered as the main reason for the worst prognosis and PCNA and Ki-67 is considered playing an important role in the process of invasion and metastasis. Recently, majority research revealed that the PCNA have a high expression in varied malignant tumors, and are associated with the invasion, metastasis and prognosis of the tumors. The aim of this study is to evaluate the expression PCNA and Ki-67 in MFH, para-tumor and normal tissue, and analyze the relationships between the expression and the clinicopathological characteristics of MFH influencing factors.Methods: Forty-eight tumor tissue samples and thirty-five tumor-surrounding tissue samples were obtained from patients with MFH undergoing radical resection, twenty-three normal tissue samples as the control. Imunohistochemistry was used to detect expression of PCNA and Ki-67. Statistical analysis was used to determine the relationship between the expression and clinicopathological parameters of MFH.Results: 1 Expression of PCNA and Ki-67 protein in MFH, para-tumor and normal tissue: PCNA-positive stain rate was 75.00%(36/48) in MFH tissue, 28.57%(10/35) in para-tuumor tissue and 21.74%(5/23) in normal tissue. Ki-67-positive stain rate was 83.33%(40/48) in MFH tissue, 25.71%(9/35) in para-tumor tissue and 21.74%(5/23) in normal tissue. Statistical analysis revealed that PCNA-positive stain rate and Ki-67-positive stain rate in MFH tissue were higher than in normal tissue(P﹤0.05) and in para-tumor tissue(P﹤0.05).2 Relationship between PCNA and Ki-67 protein expression in MFH: Both of PCNA-positive and Ki-67-positive stain was observed in 34 of 48 MFH tissue samples, PCNA-negative and Ki-67-negative stain was observed in 6 of 48 MFH tissue samples. There is a significantly correlation (P﹤0.01).3 PCNA protein expression in relation to the clinicopathological characteristics of 48 patients with MFH: PCNA-positive stain rate was 91.67%(11/12) in MFH tissue with lymph node metastasis and was 69.44%(25/36) in that without lymph node metastasis. PCNA-positive stain rate in patients with lymph node metastasis is higher than in patients without lymph node metastasis(P﹤0.01), and it is distinctly correlated with clinical stage, pathoiogical grade, tumor size and tumor recurrence(P﹤0.05),but it did not relate to age, sex, tumor location and encroachment of surrounding soft tissue (P ﹥0.05).4 Ki-67 protein expression in relation to the clinicopathological characteristics of 48 patients with MFH: Ki-67-positive stain rate was 91.7%(11/12) in MFH tissue with lymph node metastasis and was 80.56%(29/36) in that without lymph node metastasis. Ki-67-positive stain rate in patients with lymph node metastasis is higher than in patients without lymph node metastasis(P<0.01), and it is distinctly correlated with clinical stage ,pathoiogical grade, tumor size, tumor recurrence and encroachment of surrounding soft tissue(P<0.05), but it did not relate to age, sex, tumor location and tumor grade(P>0.05).5 PCNA protein, Ki-67 protein and lymph node metastasis in relation to the median survival time of 48 patients with MFH: The median survival time of 48 patients with MFH was 33 months. The median survival time was 18 months in patients with PCNA-positive stain and 34 months in patients PCNA-negative stain. The median survival time was 18 months in patients with Ki-67-positive stain but was 40 months in patients with Ki-67-negative stain respectively(P﹤0.05).Conclusions: 1 The expression of PCNA and Ki-67 in MFH tissue were both significantly higher than para-tumor tissue and than normal tissue.2 There was a significantly correlation between the expression of PCNA and Ki-67 antigen in MFH, and the expression of PCNA and Ki-67 protein may be relation to MFH lymph node metastasis. 3 The median survival time of patients with PCNA-positive stain and Ki-67-positive stain was significantly lower than that of patients with PCNA-negative stain and Ki-67-negative stain.4 Unite application of detecting PCNA antigen and Ki-67 antigen was regarded as an important clinicopathological index on judging the size of the tumor, the grade of the tumor, the malignancy of the tumor, and the prognosis of the tumor.
