Objective To investigate the role of B7-H3 in the development of clear cell renal cell carcinoma(ccRCC)and provide a novel diagnostic and potential therapeutic target for the clinical management of ccRCC.Methods From July 2003 to July 2007,76 renal cell carcinoma patients were treated by operation in our center,of these cases,69 patients who were verified as ccRCC by pathologic features were studied in this research.(1)The expression of B7-H3 in 69 excised ccRCC tissues including cancer tissues,peri-carcinoma renal tissues and normal renal tissues far away from the tumor were evaluated immunohistochemically.According to the rate and straining concentration of positive cells,we set four grades to describe the level of B7-H3 expression on ccRCC tissues:strongly positive,moderately positive,weakly positive and negative.(2)The relationship between the expression of B7-H3 and sex,age,size, pathological stage and lymph metastasis were analyzed.Results(1)The B7-H3 positive rate in the endothelial cells of the cancer tissues,cancer cells,peri-carcinoma tissues and the normal renal tissues is 79.7%,75.4%,13.0%and 0%,respectively.(2)No significant relationship was found between the expression of B7-H3 and age,sex,size and lymph metastasis.However,the patients with higher rate and concentration of B7-H3 expression have a higher pathological stage(P<0.01).Conclusion B7-H3 highly expresses in the endothelial cells of the cancer tissues and malignant cells,but seldom and none expresses in the peri-carcinoma renal tissues and the normal renal tissues far away from the masses.Based on these findings,we can conclude that B7-H3 has close relationship to the initiation and development of ccRCC.In addition,it represents a potential novel target for attacking tumor cells as well as tumor neovasculature to facilitate immunotherapeutic treatment of ccRCC tumors.
