Part 1 Polymorphisms of VISA Gene in Han Population in Northern Part of CHINA.Background Innate immunity plays an important role in the anti-virus procession.Recent years,with the deeper study of a series of Toll-like receptors,another RIG-I-Like Receptors signal conduction way has been paid more attention.RLRS was important in the process of virus recognition in most cells except pDCs.Virus-induced signaling adapter was adapter protein of RIG-I and Mda5,it activates transcription factor IRF3 through a series of signal conduction ways,activates the secretion of IFN-I and react in anti-virus procession.Objective To investigate the distribution of single nucleotide polymorphisms(SNPs)in VISA gene in ban population in northern part of china to approach its immune accommodation mechanisms.Methods(1)Objects:Healthy people of totally 124 which all come from health examination centre of Shandong Province Hospital of Shandong University.(2)Test of SNPs in VISA gene:The nucleotide sequence in flanking sequence,exon and exon-intron junction regions of VISA gene in 124 individuals were studied by DNA Sequencing technology.Nucleotide sequences were compared with Genbank sequences by nucleotide sequence analysis software.Genotypes and allele characters of SNPs were studied and compared with Caucasians.Results 6 SNPs were found in our study,in which 5 were already know,1 were newly found,and there were 9 reported sites in database that didn't appear in our study.There are different distribution sites(P<0.05)of genotype and allele frequencies between population in northern part of China and Caucasians.Conclusion(1)The results is valuable for further study of polymorphisms distribution of VISA gene in population of northern part of China.(2)To compare SNPs of population of northern part of China and Caucasians,we conclude that there exists difference between them, the results was important to explore heredity characters of VISA gene in population of northern part of China.Part 2 Relationship between Systemic Lupus Erythematosus And Polymorphisms of VISA GeneBackground Systemic lupus erythematosus(SLE)is a widespread iterate autoallergic disease.Its etiopathogenisis and pathogenesis were uncertain,maybe a combination of heredity,circumstance and hormones. Recently we presume that immunoloregulation disorder plays an important role in the process of the desease. Objective To investigate predisposing genes through the analysis of SNP distribution difference of controls and SLEs.Methods(1)Objects:SLE patients of totally 103 which all come from clinic service and hospital room of Shandong Province Hospital of Shandong University.All patients were fit with diagnosis standards revised in 1982.(2)Test of SNPs in VISA gene of SLE patients:The nucleotide sequence in flanking sequence,exon and exon-intron junction regions of VISA gene in 103 individuals were studied by DNA Sequencing technology and were compared with Genbank sequences by nucleotide sequence analysis software.Genotypes and allele characters of SNPs were studied.Results 9 SNPs were found in our study,in which 5 were already know,4 were newly found,and there were 9 reported sites in database that didn't appear in our study.There are different distribution sites(P<0.05)of genotype and allele frequencies between population in northern part of China and SLEs.Conclusion To compare SNPs of healthy people and SLEs,we can found the relationship between polymorphism sites and SLEs,and the results was profit for further study of pathogenesis of auto-immune diseases.
