【Objective】To investigate the expression ofα-B crystallin and its significance in breast cancer.To test the expression ofα-B crystallin,CD44v6,MMP-2,TIMP-2,ER,PR, HER2 and Ki67 in breast invasive ductal carcinomas,combining with tumor size, histological grading and other factors,to make a multivariate analysis with lymphonode metastasis,to explore the possibility of estimating the lymphonode metastasis and to provide the valuable information for the treatment and prognosis of breast cancer.【Methods】①Tissue microarray and immunohistochemical method were used to determine the expression ofα-B crystallin and other markers(CD44v6,MMP-2,TIMP-2, ER,PR,HER2 and Ki67)in 247 cases of breast invasive ductal carcinoma(IDC),17 cases of ductal carcinoma in situ(DCIS)and 20 cases of normal breast tissue respectively. Reverse transcriptase polymerase chain reaction(RT-PCR)was used to amplify theα-B crystallin mRNA in 17 cases of IDC.②These cases were grouped according to the clinicopathologic characteristics and molecular subtypes of breast carcinoma,and the different expressions ofα-B crystallin in individual group were evaluated.③11 potential metastatic factors were selected and analyzed by using Logistic stepwise regressin method of SPSS 12.0 software.【Results】①The different expression ofα-B crystallin between IDC(173/247, 73.04%)and DCIS(7/17,41.17%)was significant(P<0.05).②The expressions ofα-B crystallin was associated strongly with lymph node involvement(P<0.01,r=0.296).③It was significant difference within those groups of molecular subtype(P<0.05).Among them,P value is 0.007 between luminal A and HER2+ groups,0.000 between luminal A and triple-negative groups.④The adjusted value ofα-B crystallin mRNA in histologically negative lymph nodes were 0.358±0.128 in patients with IDC,but it were 0.936±0.529 in patients with lymph node involement of IDC.It had significant correlation with the lymph node metastasis(P<0.05).⑤Logistic stepwise regression analysis show that the expression ofα-B crystallin,CD44v6,MMP-2,TIMP-2 was significant related with the lymphonode metastasis.The prediction percentage of these four factors together for the lymphonode metastasis is 89.1%.【Conclusion】①The expression ofα-B crystallin is significantly higher in IDC than that in DCIS and has a positive correlation with lymph node metastasis.It also expresses higher level in groups of HER2+ and triple-negative.These suggest thatα-B crystallin may be involved in the process of progression and metastasis of the breast cancer.②The expressions ofα-B crystallin,CD44v6,MMP-2 and TIMP-2 were significant correlated with the metastasis of IDC.It is possible to estimate the metastatic potential of breast carcinomas by testing adequate metastasis-associated proteins.
